摘要(Abstract)：

目的:探讨亚慢性砷中毒对大鼠睾丸重量及组织形态的影响。方法:清洁级SD雄性大鼠40只,体重100~120 g,随机分为对照组、砷低剂量组、砷中剂量组和砷高剂量组4组,每组10只,分笼喂养;对照组自由饮蒸馏水,染砷组自由饮用含三氧化二砷(As_20_3)的蒸馏水,砷浓度分别是25、50和100 mg/L;连续喂养3个月后处死大鼠,取大鼠双侧睾丸称重并制作睾丸组织切片HE染色,采用光镜观察睾丸组织的形态改变。结果:砷中、高剂量组的睾丸重量明显降低,与对照组比较,差异具有统计学意义(P<0.05);与对照组比较,染砷组大鼠睾丸石蜡切片HE染色显示染毒组睾丸组织形态发生变化,睾丸生精小管间隙增宽,生精上皮结构疏松,层次排列紊乱并且层次逐渐减少,精原细胞出现空泡样改变,管腔中可见脱落的生精细胞,睾丸间质充血、渗出,尤其砷中、高剂量组较明显。结论:亚慢性砷中毒对睾丸组织有一定的损伤作用,这种损害作用与砷剂量可能有关。
Objective: To investigate the effect of the sub-chronic arsenic poisoning on the testis weight and tissue morphology of rats. Method: 40 Sprague-Dawley( SD) rats in cleanliness grade,weighing 100 g ~ 200 g each,were divided randomly into four groups: control group,low dose group,middle dose group and high dose group,10 in each group,and separated and fed respectively. The rats infected with sub-chronic arsenic poisoning are treated with AS2O3 dissolved in distilled water.The arsenic concentration in low,middle and high dose group is 25 mg / L,50 mg / L,and 100 mg / L,respectively. The rats in control group drink distilled water freely. The feeding continuously last three months. At the end of the experiment,all the rats were killed. The testis of both sides were weighed and testis tissue were stained with HE staining. The pathologic changes of testicle tissues were observed by light microscope. Results: The testis weight of the middle and high dose group was significantly decreased compared with that of control group,and difference was statistically significant( P < 0. 05).Compared with control group,HE staining showed that morphological changes were observed in testis of the arsenic poisoning groups,such as the gap widening in the seminiferous tubule of testis,the loose structures of the spermatogenic epithelium,disorder of arrangements of layers and gradual reductions oflayers,vacuolar changes in the spermatogonium,spermatogenic cells in the lumen,hyperemia and exudations in the interstitial testis,especially in the middle and high dose groups. Conclusion: Chronic arsenic poisoning has a certain damage to testicular tissue,which may be associated with the dosage of arsenic.
