摘要(Abstract):
目的:研究贵州苗药酢浆草水提取物的抗炎镇痛作用。方法:60只清洁级昆明系小白鼠采用冰醋酸法复制小鼠腹腔毛细血管通透性增高模型,随机分为空白组、模型组、阳性组以及酢酱草水提物低、中、高剂量组,每组10只;另取50只小鼠采用冰醋酸法复制小鼠扭体模型,随机分为模型组、阳性组以及酢酱草水提物低、中、高剂量组,每组10只;空白组和模型组给予同体积生理盐水灌胃,阳性组给予0.005 mg/g阿司匹林灌胃,酢酱草水提物低、中、高剂量组分别给予0.07 mg/g、0.14 mg/g、0.21 mg/g酢酱草水提物灌胃,均连续灌胃3 d,小鼠腹腔毛细血管通透性增高模型末次灌胃1 h后处死,用紫外分光光度计测定小鼠腹腔清洗液的吸光度值,以判定腹腔毛细血管通透性的变化;记录小鼠扭体模型10 min内的扭体次数,并计算扭体抑制率;采用DPPH实验初步考察酢酱草水提取物的自由基清除能力。结果:酢浆草水提物中、高剂量组小鼠腹腔清洗液的吸光度值低于模型组,差异有统计学意义(P<0.01或P<0.001);小鼠扭体模型10 min内扭体次数少于模型组,差异有统计学意义(P<0.001);1~100 mg/L浓度的酢浆草水提物能够发挥直接清除DPPH自由基的作用。结论:酢浆草水提物具有抗炎镇痛作用。
Objective: To investigate anti-inflammatory and analgesia effects of aqueous extracts of Miao medicine Oxalis corniculata L( CJCS). Methods: Sixty Kunming mice of clean grade were randomly divided into control group,model group,positive control group and low-dose group,middledose group,high-dose group of CJCS,10 mice per group. Among these groups,glacial acetic acid was adopted to induce and construct celiac capillary permeability model. Another fifty mice were randomly divided into model group,positive control group and low-dose group,middle-dose group,high-dose group of CJCS,10 mice per group. Among these groups,glacial acetic acid was adopted to induce and construct writhing model. Control group and model group was orally administered with equal volume of physiological saline,and positive group was orally administered with 0. 005 mg / g of aspirin. Low-dose group,middle-dose group and high-dose group was orally administered with 0. 07 mg / g,0. 14 mg / g and 0. 21 mg / g of CJCS,respectively. All groups received continuous oral administration of 3 d,and the mice were sacrificed 1 hour after the last intragastric administration of glacial acetic acid for celiac capillary permeability model. The absorbance value of celiac washing liquid of sacrificed mice was detected by UV spectrophotometer to determine changes in celiac capillary permeability. For the writhing model,the writhing times within 10 min were recorded and writhing inhibition rates were calculated.DPPH assay was adopted to preliminarily evaluate the free radical scavenging activities of aqueous extracts of CJCS. Results: The absorbance value of celiac washing liquid of middle-dose group and highdose group were significantly lower than that of model group,( P < 0. 01 and P < 0. 001). The writhing times within 10 min of middle-dose group and high-dose group were significantly lower than that of model group( P < 0. 001). The results suggested that middle-dose group and high-dose group of CJCS could remarkable suppressed the increase of peritoneal capillary permeability induced by acetic acid. 1~ 100 mg / L concentration of CJCS could show strong free radical scavenging activity in vitro. Conclusion: CJCS aqueous extracts have anti-inflammatory and analgesia effects.
