摘要(Abstract)：

目的:检测整合素β4在卵巢上皮性肿瘤中的表达水平,分析其作为卵巢肿瘤早期诊断靶标的可能性。方法:取不同病理类型卵巢上皮性肿瘤组织,Western blot检测整合素β4的表达水平。结果:相对于正常组织,整合素β4在良性卵巢上皮性肿瘤组织中升高的程度较为轻微,在该组织交界性肿瘤中的升高较良性的明显,在恶性肿瘤中的表达上调十分明显;随着分化水平的下降,整合素β4的表达相应增高;在临床早期,整合素β4的表达就有所增高,而在有转移的临床分期中,整合素β4的表达显著增加。结论:整合素β4的表达程度可以作为该类恶性肿瘤的早期诊断标志物;表达水平可以作为临床预后诊断的分子候选物。
Objective:To detect the integrin β4 expression in epithelial ovarian tumor,so as to estimate its value as a target candidate for early diagnosis of the tumor.Methods: Epithelial ovarian tumor tissues of different pathological types were ground separately in ultra low temperature caused by liquid nitrogen,and homogenized with lysis buffer for protein extraction.After running electrophoresis on SDS-PAGE,the extracted protein samples were transferred on PVDF membrane for Western blot to detect integrin β4.Results: Expression of integrin β4 increased slightly,modestly and greatly in benign tumor,borderline tumor and malignant tumor respectively compared with normal tissues.It was discovered that the expression related to differentiation and clinical stages closely.Higher expression of integrin β4 was found in poorly differentiated tumors,or in later clinical stage of the tumor.Even in the early clinical stages,the increase of integrin β4 expression could be detected,and the expression significantly increased in cases with tumor metastasis.Conclusions: In epithelial ovarian tumor tissue,the expression of integrin β4 relates to the malignancy of the tumor.Since apparent increase of integrin β4 can be found in early stage of the tumor,it is suggested that the protein can be selected as a marker for early diagnosis.The expression level of integrin β4 closely relates to the differentiation of the tumor,which indicates that integrin β4 can be used as an index for the prognosis of patients suffered from epithelial ovarian tumor.
