云贵地区淡水湖中类蛭弧菌多样性分析Diversity and Phylogenetic Analysis of Bdellovibrio-like Organisms in Fresh Water in Yun-Gui Region

摘要(Abstract):
目的:对贵州阿哈湖、百花湖及云南滇池不同深度湖水中分离的类蛭弧菌进行分类鉴定,了解云贵地区淡水中类蛭弧菌多样性。方法:培养云贵地区3个湖泊中分离的类蛭弧菌,进行16S rRNA基因测序分析,用MEGA 6.0软件以邻接法构建3个湖泊中类蛭弧菌16S rRNA序列的系统发育树,并以最大似然法(PHYLIP3.1软件)加以验证。结果:从阿哈湖、百花湖及滇池不同深度湖水中分离的类蛭弧菌覆盖率C值为75%,根据类蛭弧菌16S rRNA构建发育树,本研究所得的类蛭弧菌分属3个簇群,γ-Proteobacteria(8.3%)、δ-Proteobacteria(50.0%)及Sphingobacteria(41.7%);在百花湖中分离得到1株路德维希肠杆菌,分离自贵州阿哈湖的6株菌株与噬菌弧菌属的Bacteriovorax sp.EPA同源性较高,分离于滇池的5株菌株的序列为不可培养细菌序列。结论:云贵地区阿哈湖、百花湖及滇池不同深度湖水中的类蛭弧菌具有较高多样性,推测在云南滇池区域存在着尚未开发的新型类蛭弧菌资源。
Objective: To identify Bdellovibrio-like organisms( BALOs) collected from Aha lake and Baihua lake in Guizhou,and from different depths of Dianchi lake in Yunnan,and to study their diversity in fresh water of Yun-Gui region. Method: Sequences of 16 S rRNAs of collected BALOs were submitted to international molecular data bank( GenBank) for similarity,and phylogenetic trees were constructed to analyze the phylogeny with MEGA( version 6. 0). Results: The coverage C value of these BALOs was estimated 75%; The Phylogenetic trees showed that the majority belonged to 3 clusters: γ-Proteobacteria( 8. 3%),δ-Proteobacteria( 50. 0%),and Sphingobacteria( 41. 7%); One strain of Enterobacter ludwigii was isolated from Baihua lake,and other 6 strains from Aha lake showed high homology with Bacteriovorax sp. EPA. However,5 strains from Dianchi lake in Yunnan were failed to be identified at genus level. Conclusions: There is a high diversity of BALOs in Aha lake and Baihua lake in Guizhou,and in different depths of Dianchi lake in Yunnan. It is inferred that there might be new types of BALOA resources in fresh water of Dianchi lake waiting for exploiting.
