环境因素对大鼠缺血心肌细胞凋亡及Bcl-2、Bax、Caspase-3表达的影响

摘要(Abstract):

目的:探讨光照、黑暗、行为限制对心肌梗死大鼠缺血心肌细胞凋亡及Bcl-2、Bax、Caspase-3表达的影响。方法:Wistar大鼠随机分为假手术组(A组)、心肌梗死组(B组)、黑暗组(C组)、光照组(D组)及行为限制组(E组),用TUNEL法检测心肌凋亡细胞,免疫组织化学和逆转录-聚合酶链反应(RT-PCR)方法分别检测Bcl-2、Bax、Caspase-3的蛋白及基因表达。结果:(1)与A组比较,B、C、D、E组的凋亡指数增加(P<0.01);与B组比较,D、E组的凋亡指数增加(P<0.05);与C组比较,D、E组的凋亡指数增加(P<0.05)。(2)与A组比较,B、C、D、E组的Bcl-2、Bax、Caspase-3蛋白表达增加(P<0.01);与B组比较,D、E组的Bax、Caspase-3蛋白表达增加(P<0.05);与C组比较,D、E组的Bax、Caspase-3蛋白表达增加(P<0.05)。(3)与A组比较,B、C、D、E组的Bcl-2、Bax、Caspase-3 mRNA表达增加(P<0.01);与B组比较,D、E组的Bax、Caspase-3 mRNA表达增加(P<0.01);与C组比较,D、E组的Bax、Caspase-3 mRNA表达增加(P<0.01)。结论:光照环境、行为限制可增加大鼠缺血心肌细胞凋亡,其机制可能与上调Bax、Caspase-3的表达有关。
Objective: To study the effects of dark,lighting and behavior limitation on ischemic cardiac myocyte apoptosis and expression of Bcl-2,Bax and Caspase-3 in rats with myocardial infarction(AMI).Methods: Forty Wistar rats were randomly divided into five groups with 8 in each group: sham-operation group(group A),AMI group(group B),dark group(group C),lighting group(group D),and behavior limitation group(group E).Apoptosis of cardiac myocyte was detected with TUNEL method,and the expression of Bcl-2,Bax and caspase-3 were examined with immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR).Results:(1) Apoptosis indexes of groups B,C,D and E were higher than those of group A(P<0.01).Apoptosis indexes of groups D and E were higher than those of group B(P<0.05).Apoptosis indexes of groups D and E were higher than those of group C(P<0.05).(2) The expression of Bcl-2,Bax and Caspase-3 protein in groups B,C,D and E were higher than those of group A(P<0.01).The expression of Bax and Caspase-3 protein of groups D and E were higher than those of group B(P<0.05).The expression of Bax and Caspase-3 protein of groups D and E were higher than those of group C(P<0.05).(3) The expression of Bcl-2,Bax and Caspase-3 mRNA in groups B,C,D and E were higher than those of group A(P<0.01).The expression of Bax and Caspase-3 mRNA of groups D and E were higher than those of group B(P<0.01).The expression of Bax and Caspase-3 mRNA of groups D and E were higher than those of group C(P<0.01).Conclusions: Lingting,behavior restriction could increase ischemic cardiac myocyte apoptosis in rats.The mechanisms may be associated with up-regulating the expression of Bax and Caspase-3.Dark environment does not increase cardiac myocyte apoptosis in rats.
