G-MDSCs及HO-1对异基因造血干细胞移植后患者发生急性移植物抗宿主病的预测价值
The predictive value of G-MDSCs and HO-1 for aGvHD in patients after allogeneic hematopoietic stem cell transplantation

摘要(Abstract):

目的 探讨粒细胞的髓系来源抑制性细胞(G-MDSCs)和血红素加氧酶-1(HO-1)对异基因造血干细胞移植(allo-HSCT)患者发生急性移植物抗宿主病(aGvHD)的预测价值。方法 收集29例allo-HSCT术患者的一般临床资料,据1974年西雅图分级标准判断aGvHD发生情况分为有aGvHD组和无aGvHD组,抽取2组患者移植后第2、3、4、6、8周及第3月时全血,运用多色流式细胞术(FCM)检测患者外周血G-MDSCs及胞内HO-1和白细胞介素-10(IL-10)的水平,采用表面抗原染色检测G-MDSCs的百分比和绝对值计数,采用破膜染色检测G-MDSCs中HO-1和IL-10的中位荧光强度(MFI)、表达百分比及绝对值计数。结果 2组患者allo-HSCT后第2周于外周血中均可检测到G-MDSCs,但第4周时有aGvHD组患者外周血G-MDSCs较无aGvHD组升高(P<0.05);破膜染色结果显示,有aGvHD组患者于allo-HSCT后第4周时G-MDSCs内HO-1高于无aGvHD组(P<0.05)。结论 allo-HSCT患者术后第4周外周血中G-MDSCs和HO-1的高表达可预测aGvHD的发生。
Objective To explore the predictive value of myeloid derived inhibitory cells(G-MDSCs) and heme oxygenase-1(HO-1) of granulocytes in patients with acute graft versus host disease(aGvHD) who underwent allogeneic hematopoietic stem cell transplantation(allo-HSCT). Methods The general clinical datas of 29 patients with aGvHD who underwent allo-HSCT were collected. According to the Seattle criteria in 1974 and the occurrence of aGvHD, the patients were divided into two groups: aGvHD group and non-aGvHD group. The whole blood samples were collected in the 2~(nd), 3~(rd), 4~(th), 6~(th), 8~(th) weeks and the 3~(rd) months after patients' transplantation. The expression of G-MDSCs and intracellular HO-1 and IL-10 in peripheral blood of the two groups were detected by multi-color flow cytometry(FCM). Concretely, the percentage and absolute count of G-MDSCs were detected by surface antigen staining. In addition, the median fluorescence intensity(MFI), percentage and absolute count of HO-1 and IL-10 in G-MDSCs were detected by membrane-breaking staining. Results G-MDSCs could be detected in peripheral blood of patients in the 2~(nd) week after allo-HSCT. In the 4~(th) week, G-MDSCs in aGvHD group were higher than those in non-aGvHD group(P<0.05). The results of membrane-breaking staining showed that HO-1 in G-MDSCs was higher in aGvHD group than in non-aGvHD group in the 4~(th) week after allo-HSCT(P<0.05). Conclusion The high expressions of G-MDSCs and HO-1 in peripheral blood of the patients in the 4~(th )week after allo-HSCT can predict the occurrence of aGvHD.
