DC联合CIK治疗对肝癌模型大鼠免疫功能的影响

摘要(Abstract):

目的 探讨树突状细胞(DC)联合细胞因子诱导的杀伤细胞(CIK)治疗对肝癌模型大鼠免疫功能的影响。方法 采用H-35肿瘤细胞在Wistar大鼠上建立肝癌模型,分为对照组、CIK治疗组和DC+CIK治疗组,治疗组大鼠接受自体移植的CIK或(和)DC细胞;治疗后分别于1、4、7、10及13 d测量肿瘤大小,于第14天处死大鼠前收集血清用于检测谷草转氨酶、丙氨酸转移酶、总胆红素及白蛋白等肝功能指标,通过ELISA实验检测血清γ-干扰素(IFN-γ)、白细胞介素4(IL-4)及肿瘤坏死因子-α(TNF-α)水平,实时荧光定量PCR和蛋白质印迹法检测B淋巴细胞瘤-2基因(Bcl-2)和B淋巴细胞瘤-2关联X(Bax)的mRNA及蛋白表达水平。结果 CIK治疗组和DC+CIK治疗组大鼠的肿瘤均小于对照组,大鼠的肝功能得到改善,IFN-γ、IL-4和TNF-α的表达量增加,Bcl-2的表达降低,Bax的表达增加,差异均有统计学意义(P<0.05);DC+CIK联合治疗组大鼠的疗效明显优于CIK治疗组,差异有统计学意义(P<0.05)。结论 DC+CIK联合疗法可能通过调节机体细胞因子表达水平发挥抗肿瘤的作用,且疗效优于单纯CIK治疗。
Objective To investigate the effect of dendritic cells( DC) combined with cytokine induced killer cell(CIK) treatment on immune function in rat liver cancer model.Methods Wistar rats were implanted with RH-35 liver cancer cells to establish liver cancer model. These liver cancerbearing rats were divided into control group, CIK treatment group and DC + CIK treatment group. CIK or DC cells were transplanted into the treatment group. Tumor sizes were measured at 1, 4, 7, 10,and 13 days after the treatment. Serum was collected before sacrificing rats on the 14thday to detect glutamic oxaloacetic transaminase, alanine transferase, total bilirubin and albumin. The serum levels of IFN-γ, IL-4, and TNF-α were detected by ELISA assays. The mRNA and protein expressions of B cell lymphoma-2(Bcl-2) and B cell lymphoma/leukemia-2(Bax) were detected by real-time PCR and Western blot, respectively.Results Both CIK and DC + CIK treatment significantly reduced tumor sizes and improved liver function, increased secretion of IFN-γ, IL-4, and TNF-α, decreased Bcl-2 expression and enhanced Bax expression(P< 0. 05 compared to control group). DC + CIK combined therapy was significantly better than CIK therapy.Conclusion The combination therapy of DC + CIK may exert an anti-tumor effect via regulating the expression level of serum cytokines, and its therapeutic effect is better than that of CIK alone.
