摘要(Abstract):

目的:探讨长链非编码RNA SNHG16对胃癌细胞AGS增殖调控的影响。方法:AGS细胞分control组(正常培养的AGS细胞,C组)、negative control组(转染si-NC序列,NC组)及si-SNHG16组(转染si-SNHG16序列,SI组),通过siRNA干扰技术敲低lncRNA SNHG16后,采用逆转录-实时荧光定量PCR法(RT-qPCR)验证AGS细胞中SNHG16的表达水平,CCK-8法检测敲低lncRNA SNHG16后AGS细胞增殖能力的改变,流式细胞术检测AGS细胞周期的改变,Western blot检测AGS细胞周期蛋白D1(cyclin D1)、周期蛋白依赖性激酶6(CDK6)及细胞周期蛋白依靠性激酶抑制剂家族成员p21蛋白表达水平的改变。结果:敲低lncRNA SNHG16后,与NC组相比,SI组中lncRNA SNHG16的表达显著降低(P <0. 01),细胞增殖能力受到明显抑制,细胞周期阻滞在G0/G1期,细胞周期相关蛋白cyclin D1及CDK6表达下调,p21明显上调(P均<0. 01)。结论:敲低lncRNA SNHG16可能通过上调p21影响胃癌细胞AGS的增殖。
Objective: To investigate the effect of long non-coding RNA small nucleolar RNA host gene 16( LncRNA SNHG16) on gastric cancer cell AGS proliferation. Methods: AGS cell were divided into control group( conventional cultivated AGS cell,group C),negative control group( transfected si-NC sequence,group NC) and si-SNHG16 group( transfected si-SNHG16 sequencde,group SI).After knockdown of lncRNA SNHG16 by siRNA interference,the expression level of lncRNA SNHG16 was verified by RT-qPCR,the proliferation ability of AGS cell was evaluated by CCK-8 assay. Cell cycle progression was measured by flow cytometric analysis; Western blot was performed to detect the expression of proteins as cyclinD1,CDK6 and p21. Results: Compared with NC group,the expression of lncRNA SNHG16 of SI group was significantly decreased( P < 0. 01) and the cell proliferation ability was remarkably inhibited after knockdown of lncRNA SNHG16. Furthermore,cell cycle was arrested in G0/G1 phase. The expressions of cell cycle-related proteins( cyclinD1,CDK6) were down-regulated and p21 was observably up-regulated( P < 0. 01). Conclusion: Knockdown of lncRNA SNHG16 may affect the proliferation of gastric cancer cell AGS via up-regulating p21.
