摘要(Abstract):
目的:探讨h FAM92A1在人宫颈癌He La细胞不同周期时相的表达差异。方法:常规培养Hela细胞,应用血清饥饿法将He La细胞同步化于G1期,胸腺嘧啶核苷双阻断法同步化于S、G2期,秋水仙素阻断法同步化于M期,应用流式细胞术检测同步化效率,应用RT-PCR方法检测h FAM92A1在人宫颈癌He La细胞不同周期时相的表达水平。结果:流式细胞仪检测细胞同步化效率G1期为77.5%、S期为85.5%、G2期为61.9%、M期为44%;FAM92A1基因在各期均有表达,但存在差异,以S期表达最高。结论:FAM92A1基因在He La细胞不同周期时相的表达有较大差异。
Objective: To investigate the expression differences of h FAM92A1 in different cell cycle time phases of He La cells of human cervical carcinoma. Methods: He La cells were conventionally cultured and synchronized in G1 phase using serum starvation,synchronized in G2 and S phase using double thymidine blocking,synchronized in M phase using colchicine blocking,and synchronizing efficiency was tested by flow cytometry. RT-PCR method was adopted to detect the expression of h FAM92A1 in different cell cycle phases of He La cells. Results: Flow cytometry instrument detected cell synchronization efficiency as follows: G1 phase 77. 5%,S phase 85. 5%,G2 phase 61. 9%,M phase 44%. FAM92A1 genes were expressed in each period but with differences,the highest expression was in S phase. Conclusions: FAM92A1 gene of He La cell expression in different cycle phases exhibit notable differences.
