摘要(Abstract):
目的:观察子宫内膜干细胞(En SCs)来源细胞因子"鸡尾酒"(EdCC)对乳小鼠心肌细胞氧化应激损伤和凋亡的保护作用。方法:乳小鼠心肌细胞分为正常组、对照组和EdCC组,用过氧化氢(H_2O_2)造成氧化应激,检测上清液乳酸脱氢酶(LDH)活性,TUNEL染色检测细胞凋亡,DCFH-DA检测细胞内活性氧族(ROS)浓度。结果:En SCs呈成纤维细胞样贴壁生长,EdCC蛋白提取效率为每24 h提取(241.3±32.1)μg/10~6个细胞;与对照组比较,EdCC组(100 mg/L)上清液LDH活性明显降低,心肌细胞凋亡率明显降低,细胞内ROS浓度降低,差异均有统计学意义(P<0.01)。结论:EdCC减轻体外培养乳小鼠心肌细胞氧化损伤及凋亡,可能与降低细胞内ROS水平有关。
Objective: To investigate the protective effect of endometrial stem cells( En SCs) derived from cytokine cocktail( EdCC) on neonatal mouse cardiomyocytes under oxidative stress induced injury and apoptosis. Methods: Neonatal mouse cardiomyocytes were divided into normal group,control group and EdCC group. H_2O_2 was used to induce oxidative stress. The lactate dehydrogenase( LDH)activity in supernatants was determined to evaluate cell damage,TUNEL staining was used to count apoptotic cells and DCFH-DA was used to measure intracellular reactive oxygen species( ROS) concentration. Results: En SCs were adherent fibroblast-like cells and the efficiency of EdCC concentration was( 241. 3 ± 32. 1) g /10~6 cells in every 24 h. As compared with control group,the LDH activity in supernatants greatly reduced in EdCC group( EdCC 100 g / m L)( P < 0. 01),and also the apoptotic cardiomyocyte ratio( P < 0. 01) and intracellular ROS concentration( P < 0. 01),difference was statistical significant. Conclusion: EdCC alleviated oxidative stress induced injury and cell apoptosis in neonatal mouse cardiomyocytes,possibly associated with decreasing intracellular ROS level.
