肝癌模型大鼠癌组织组蛋白乙酰化水平观察Observation of Histone Acetylated Levels in Hepatoma Tissues of Rat Models

摘要(Abstract):
目的:观察肝癌模型大鼠癌组织中组蛋白H3K9、H4K12、H4K16位点的乙酰化水平。方法:50只大鼠随机分为模型组(40只)及对照组(10只),予模型组大鼠2.5 g/L二乙基亚硝胺(DEN)溶液自由饮用14周建立肝癌模型,对照组大鼠自由饮用灭菌食用水;造模结束后,取肝组织镜下观察,确认造模成功,采用免疫组织化学及Western blot检测肝癌组织中组蛋白H3K9、H4K12及H4K16乙酰化水平。结果:造模过程中模型组大鼠死亡20只,另20只大鼠肝组织肝小叶结构破坏,假小叶形成,在11个标本中可见肝癌细胞;免疫组织化学染色及Western Blot检测结果显示,模型大鼠肝癌组织组蛋白H3K9、H4K12及H4K16位点乙酰化水平均低于对照组大鼠肝组织(P<0.05)。结论:DEN诱导的肝癌模型大鼠肝癌组织组蛋白H3K9、H4K12及H4K16位点的乙酰化水平均明显降低,提示组蛋白H3K9、H4K12、H4K16位点的低乙酰化可能参与了肝癌的发生。
Objective: To observe the acetylated levels of histone H3 K9,H4 K12 and H4 K16 in hepatocellular carcinoma cells( HCC),so as to establish a foundation for further research of the mechanism of hepatoma. Method: 40 rats were induced by administering 2. 5 g/L diethylnitrosamine water as the model group,while 10 rats were treated with sterile edible water as the normal controls. The general condition of rats was observed during modeling. After modeling,the rats were weighed and sacrificed after anesthesia. Their livers were was observed by the eye and microscopic. Once hepatoma was identified by microscopic,the acetylated levels of histone H3 K9,H4 K12 and H4 K16 in hepatoma tissues of the rats were detected by western blot experiment and immuno-histochemistry. Result: During modeling,rats in the model group were gradually lethargic. They lost thiir body luster,appetite and weight and some rats died,but rats in the control group were generally in good condition. Visible to the naked eyes: the volume of liver tissues in model group increased and surface was covered with several nodules of different sizes,hemorrhage and necrosis,but the liver tissues in the control group were normal. Visible to microscopic: the structure of the hepatic lobule was normal in the control group; the hepatic lobule structure of the model group was damaged and the false lobular formation was formed. In addition,HCC were found in 11 specimens. Compared with normal liver tissues,the acetylated level of histone H3 K9,H4 K12 and H4 K16 in rat hepatoma tissues was lower. Conclusion: The acetylated level of histone H3 K9,H4 K12 and H4 K16 have associations with the occurrence of hepatoma.
