丹参酮Ⅱ-A对大鼠成骨细胞体外增殖分化的影响

摘要(Abstract):

目的:研究丹参酮Ⅱ-A(Ts II-A)对大鼠成骨细胞(OB)体外增殖分化的影响。方法:分离、培养、纯化SD乳鼠颅盖骨OB,选择传代至第3代的OB进行细胞形态学鉴定;将第3代OB分为实验组及对照组,实验组给予5×10-3mg/L、5×10-2mg/L及5×10-1mg/L Ts II-A处理,对照组OB加入与实验组等量的完全培养基DMEM;于接种后第1、3、5、7天4个时间点观察Ts II-A对OB增殖的影响;于接种后第5天检测OB中碱性磷酸酶(ALP)活性。结果:分离、培养所得的细胞符合OB的形态学特征;与对照组比较,实验组不同浓度Ts II-A对OB均有促增殖的作用(P<0.05),其中5×10-2mg/L浓度组促增殖作用最大;培养OB至第5天时实验组中ALP活性高于对照组(P<0.05)。结论:Ts II-A在一定的作用时间内,对体外培养的OB具有促增殖分化作用。
Objective: To investigate the effect of Tanshinone Ⅱ-A( Ts II-A) on the proliferation and differentiation of rat osteoblast( OB) in vitro. Methods: Infant rats' calvarial OB were isolated,cultured and purified in vitro,and passed to the third generation. The morphological changes and physiologic characters of the third generation OB were observed by inverted microscope. The third generation OB were divided into experimental group and control group. The experimental group was treated by different concentrations of Ts II-A while control group were given equal amounts of pure DMEM culture medium. Then the effects of Ts II-A on the proliferation and differentiation of rats' OB on the 1stday,the 3rdday,the 5thday,and the 7thday,were observed respectively. The alkaline phosphatase( ALP)activities of OB were determined 5 days after inoculation. Results: The morphological characters of isolated and cultured cells accorded with those of OB. Different concentrations of Ts II-AT had significant promotion effects on OB proliferation,of which the 5 × 10- 2mg / L of Ts II-AT showed the maximum promotion effect( P < 0. 05). The ALP activities of experimental group were significantly higher than those of control group 5 days after inoculation( P < 0. 05). Conclusion: Ts II-A has the promotion effect on OB proliferation and differentiation in vitro within certain period of time.
