维生素C对心肌缺血/再灌注家兔心功能和SOD、MDA的影响
Effects of Vitamin C on Superoxide Dismutase and Malondialdehyde in Myocardium and Cardiac Function of Rabbits during Ischemia/Reperfusion

摘要(Abstract):

目的:观察维生素C(VitC)对心肌缺血/再灌注(I/R)家兔心功能和心肌组织中超氧化物歧化酶(SOD)和丙二醛(MDA)的影响。方法:家兔24只随机分为3组,假手术组(A组)、心肌缺血/再灌注组(B组)、VitC组(C组);用生物机能实验系统记录各组家兔左室收缩峰压(LVSP)、左室舒张末压(LVEDP)和左室内压最大收缩/舒张变化速率(±dp/dtmax);用黄嘌呤氧化酶法测定SOD,硫代巴比妥酸法测定MDA含量。结果:与A组比较,B、C两组的±dp/dtmax减低,有显著性差异(P<0.01);与B组比较,C组±dp/dtmax增高,有统计学差异(P<0.05);与A组比较,B、C两组心肌组织SOD活性减低,而MDA含量增高(P<0.05或P<0.01);与B组比较,C组心肌组织中SOD活性增高,而MDA含量降低(P<0.05)。结论:VitC能够改善心肌I/R家兔的心功能,其机制可能与抑制机体内脂质过氧化有关。
Objective: To investigate the effects of vitamin C(VC) on contents of superoxide dismutase(SOD),and malondialdehyde(MDA) in myocardium and cardiac function of rabbits during ischemia/reperfusion(I/R);Methods: Twenty four rabbits were divided randomly into 3 groups(8 in each): sham-operated group(group A),I/R group(group B),and VC treated group(group C).Organism function experiment system was used for recording left ventricular systolic pressure(LVSP),left ventricular end diastolic pressure(LVEDP),and maximal changing rate of left ventricular inner pressure(±dp/dtmax);SOD in myocardium of rabbits was detected with xanthine oxcydation method and MDA detected with thiobarbituric acid method.Results: ± dp /dtmax in groups B and C were lower than that in group A(P<0.01),but ± dp /dtmax in group C was higher than that in group B(P<0.05 or P<0.05).SOD in myocardium was lower and MDA was higher in group B and group C than those in group A(P<0.01),while SOD content was higher and MDA was lower obviously in group C than those in group B(P<0.05).Conclusions: VC could improve cardiac function in rabbits during I/R.The mechanism might relate to reduction of over-oxidative reaction of lipid.
