枸杞多糖对甲醛所致雄性小鼠生殖毒性的拮抗作用
Study on the Antagonism of Lycium Barbarum Polysaccharides on Formaldehyde Induced Male Mice Reproductive Toxicity

摘要(Abstract):

目的:观察枸杞多糖(LBP)对甲醛(FA)所致雄性小鼠生殖毒性的拮抗作用。方法:将96只健康雄性昆明种小鼠随机分为FA低、中、高剂量染毒组,LBP低、中、高剂量拮抗组,阳性对照组和阴性对照组;染毒组和阴性对照组小鼠分别给予0.2、2.0及20 mg/(kg.d)的FA和生理盐水腹腔注射,1次/d,连续14 d;阳性对照组小鼠给予50 mg/(kg.d)环磷酰胺腹腔注射,隔天1次,共14 d;LBP拮抗组小鼠腹腔注射20 mg/(kg·d)FA同时,分别给予50、100及200 mg/(kg.d)的LBP溶液灌胃,1次/d,连续14 d;给药结束后,处死小鼠,计算小鼠附睾、睾丸脏器系数,显微镜下计数各组小鼠精子数量、活动率及畸形率,采用化学发光法检测小鼠血清中的睾酮(T)水平。结果:与阴性对照相比较,FA低、中、高剂量染毒组睾丸脏器系数、精子计数、活动率和血清中T含量显著减低,精子畸形率显著增加,差异有统计学意义(P<0.01);与FA高剂量组相比较,LBP低、中、高剂量拮抗组睾丸脏器系数、精子计数、活动率和血清中T含量显著增加,精子畸形率显著减低,差异有统计学意义(P<0.01)。结论:FA染毒对雄性小鼠有生殖毒性作用,LBP可拮抗FA致雄性小鼠生殖毒性的作用。
Objective: To observe effects of antagonism of Lycium Barbarum Polysaccharide( LBP)on formaldehyde induced reproductive toxicity of male mice. Methods: 96 male Kunming mice were divided randomly into low,medium and high dose formaldehyde groups; low,medium and high antagonism groups of LBP,positive control group and negative control group. Groups of formaldehyde and negative control groups were injected with formaldehyde with the concentration of 0. 2,2. 0 and 20 mg/( kg. d) and saline intraperitoneal injection once a day for 14 days continuously; positive control group was injected with 50mg/( kg. d) cyclophosphamide by intraperitoneal injection once every other day for14 days; antagonism groups of LBP were injected with 20 mg/( kg. d) formaldehyde by intraperitoneal injection,and were given 50,100 and 200 mg/( kg. d) LBP by gavage once a day for 14 days continuously; after medication,mice were put to death and calculating mice testis,epididymis,counted the number of sperm,mobility and deformity by microscope counting method; chemiluminescence method was adopted to detect serum level of testosterone( T) in mice. Results: Comparing with the negative control group,formaldehyde low,medium and high dose group infected testicular viscera coefficient,sperm count,the activity rate and significantly reduce T content in serum and sperm deformity rate increased significantly( P < 0. 01) testis organ coefficients; comparing with formaldehyde high dose group,LBP low,medium and high dose antagonist group of testicular viscera coefficient,sperm count,T content in serum and activity rate increased significantly,the sperm malformation rate was significantly lower( P < 0. 01). Conclusions: formaldehyde contanminated male mice reproductive toxicity;LBP could antagonize the male reproductive toxicity of formaldehyde.
