头花蓼对幽门螺杆菌生长及代谢相关基因的影响

摘要(Abstract):

目的:观察苗药头花蓼对幽门螺杆菌(H.pylori)生长代谢及其相关基因的表达的影响。方法:将H.pylori菌接种于不同浓度的头花蓼培养基(头花蓼终浓度分别为64、32、16、8、4、2、1及0.5 g/L),测定无H.pylori菌生长时头花蓼的最小抑菌浓度(MIC);将H.pylori菌接种于无头花蓼培养基(对照组)和头花蓼MIC培养基(头花蓼组)培养36 h,每隔4 h采用用酶标仪检测590 nm处的吸光度值,绘制H.pylori生长曲线;用实时荧光定量PCR(Real-Time PCR)检测2组H.pylori菌液生长代谢相关基因Frd A、Frd B、Hyu A及Pfr的表达水平。结果:头花蓼对H.pylori最低抑菌浓度MIC为4 g/L,头花蓼组H.pylori生长曲线位于对照组之下,Real-time PCR结果显示头花蓼作用后,头花蓼组生长代谢相关基因Frd A、Frd B、Hyu A及Pfr基因m RNA均呈减弱趋势,差异具有统计学意义(P<0.05)。结论:头花蓼在体外对H.pylori具有明显抑制作用,可通过影响细菌生长代谢相关基因的表达来抑制H.pylori的生长。
Objective: To observe the effect of Polygonum capitatum on growth metabolism associated genes expression of H. pylori. Methods: H. pylori fungus was inoculated in varied Polygonum capitatum concentration medium( final concentration as 64,32,16,8,4,2,1 and 0. 5 g / L); testing minimal inhibitory concentration( MIC) of Polygonum capitatum when no H. pylori grows; H. pylori fungus was inoculated in no- Polygonum capitatum( control group) and Polygonum capitatum MIC( Polygonum capitatum group) to cultivate for 36 h,and test Absorbance value of 590 nm in every 4 h by ELIASA; then draw the H. pylori growth curve; real-time fluorescence quantitative PCR was adopted to test expression level of growth metabolism associated genes Frd A,Frd B,Hyu A and Pfr both groups H. pylori fungus solution. Results: the min MIC concentration of Polygomun capitutam on H. pylori was 4 g /L,growth curve of Polygomun capitutam H. pylori was below control group; Real-time PCR results showed that after Polygomun capitutam taking effect,growth metabolism associated genes of Polygomun capitutam group,m RNA of Frd A,Frd B,Hyu A and Pfr gene were decreasing,differences had statistical significance( P < 0. 05). Conclusion: Polygonum capitatum has obvious inhibitory effect on H. pylori in vitro,Polygonum capitatum can inhibit the growth of H. pylori by affecting the growth of growth metabolism related genes.
