白头翁皂苷B4在Calu-3细胞单层中的渗透性研究

摘要(Abstract):

目的:建立白头翁皂苷B4的HPLC测定方法,研究其在单层Calu-3细胞中的渗透性。方法:色谱条件为Agilent C18柱(4.6 mm×250 mm,5μm)、流动相为0.1%磷酸-乙腈(71%~29%)、流速为0.8 m L/min、检测波长205 nm、柱温25℃,考察不同浓度B4在Calu-3单层细胞中的渗透情况,以卡波姆974P为促渗剂,考察卡波姆对B4渗透性的影响。结果:B4在1.5~30.0 mg/L内与峰面积呈良好线性关系(r=0.999 3),B4在Calu-3细胞的渗透性具有浓度依赖性,卡波姆的加入提高了B4的渗透量。结论:B4的渗透以细胞旁路为主要途径,而卡波姆的加入可提高细胞旁路通透性,使药物渗透量增加。
Objective: To establish a HPLC method to investigate of anemoside B4 permeability in Calu-3 cell monolayer. Methods: The samples were analyzed with a Agilent C18 column( 4. 6 mm ×250 mm,5 μm) at 25 ℃. The mobile phase was water containing 0. 1% H3 PO4-acetonitrile( 71% ∶29%). The flow rate was 0. 8 m L/min. The transport of B4 in Calu-3 monolayer cells was investigated with different concentrations of B4,and Carbopol 974 P was used as a penetration enhancer to determine the effect of carbopol on B4 permeability. Results: The line-arity of B4 calibration curve was good in the range of 1. 5 ~ 30. 0 mg/L( r = 0. 999 3). The minimal quantitation limit was 1. 5 mg/L.The penetration of B4 on Calu-3 cells was concentration-dependent,and the addition of Carbopol increased the penetration of B4. Conclusions: The absorption of B4 on mucous membrane is mainly through paracellular pathway,and the Carbopol can enhance the permeation of B4.
