丁苯酞对大鼠窒息性心脏骤停心肺复苏后全脑损伤的保护作用

摘要(Abstract):

目的:探讨丁苯酞对大鼠窒息性心脏骤停心肺复苏后全脑损伤的保护作用。方法:健康SD大鼠45只随机分为假手术组、心脏骤停复苏组及丁苯酞干预组,后两组制作窒息性心脏骤停心肺复苏模型,于心肺复苏后自主循环恢复6、12、24、48及72 h时分别处死大鼠3只,取脑组织测量一氧化氮(NO),测定血清神经元特异性烯醇化酶(NSE)、细胞色素C(Cyt-C)、高迁移率蛋白B1(HMGB1)及脑组织含水量。结果:心脏骤停复苏组和丁苯酞干预组大鼠自主循环恢复后脑组织NO、血清NSE、Cyt-C、HMGB1及脑组织含水量均呈现出增加态势,24 h达到最高点,然后出现降低,直到72 h;心脏骤停复苏组和丁苯酞干预组大鼠心肺复苏后自主循环恢复各时间点脑组织NO、血清NSE、Cyt-C、HMGB1及脑组织含水量均明显高于相应时间点假手术组大鼠(P<0.05);丁苯酞干预组大鼠心肺复苏自主循环恢复后各时间点脑组织NO、血清NSE、Cyt-C、HMGB1及脑组织含水量均明显低于相应时间点心脏骤停复苏组大鼠(P<0.05)。结论:丁苯酞可减轻大鼠窒息性心脏骤停心肺复苏后全脑损伤,具有一定的保护作用。
Objective:To investigate the protective effect of butylphthalide on whole brain injury in cardiac arrest rats after cardiopulmonary resuscitation. Methods:Forty-five healthy SD rats,body weighted 200 ~ 250 g,were divided into 3 groups randomly( 15 in each) :sham group( group S),cardiac arrest resuscitation group( group C),and butylphthalide intervention group( group B). In6 h,12h,24 h,48h,and 72 h of spontaneous circulation recovery time after cardiopulmonary resuscitation 3 rats were killed in each group,and nitric oxide( NO),neuron specific enzyme( NSE),cytochrome C( Cyt-C),high mobility protein B1( HMGB1),and water content in brain tissue were detected. Results:NO,serum NSE,Cyt-C,HMGB1,and brain water content at 6h,12 h,24h,48 h,and72h after cardiopulmonary resuscitation in groups C and B were significantly higher than those in group S in responding time points( P < 0. 05),and showed an increasing tendency from beginning,and reached the highest point at 24 h,and then decreased,until to 72 h. NO,serum NSE,Cyt-C,HMGB1,and brain water content at different time points after cardiopulmonary resuscitation in group B were significantly lower than those in group C( P < 0. 05). Conclusion:Butylphthalide can relieve whole brain injury in cardiac arrest rats after cardiopulmonary resuscitation,which has certain protective effect.
