糖尿病小鼠肾组织中细胞焦亡发生的现象研究

摘要(Abstract):

目的:观察糖尿病(DM)小鼠肾组织中是否发生细胞焦亡及相关蛋白表达变化。方法:以链脲佐菌素(STZ)复制DM C57BL/6J小鼠模型(DM组),并设立正常对照组(NC组); 16周后处死小鼠,测量血清中血糖和血肌酐,采用HE、Masson染色观察两组小鼠肾组织的变化,应用Western blot法及Real Time-PCR检测活性半胱天冬氨酸蛋白酶-1(Caspase-1)、细胞焦亡关键蛋白Gasdermin-D(GSDMD)及其mRNA表达水平。结果:与NC组相比,DM组小鼠血糖及血肌酐升高,HE及Masson染色下见DM组小鼠肾组织中肾小球内基质沉积和胶原纤维增生,部分肾小管上皮细胞可见细胞肿胀、空泡变性、管腔扩张及基底膜增厚; DM组中活性Caspase-1、GSDMD的蛋白及mRNA表达较NC组增高,差异具有统计学意义(P <0. 05)。结论:DM小鼠肾组织中可能发生由Caspase-1激活介导GSDMD触发的细胞焦亡。
Objective: To observe the pyroptosis and changes of related protein expression in kidney tissues of diabetic mice( DM). Methods: The C57 BL/6 J DM model was replicated as DM group with streptozotocin( STZ),and the normal control group( NC group) was established. 16 weeks later the mice were sacrificed,and the biochemical parameters were analyzed. The kidney tissues were sampled for HE and Masson staining to observe the pathological changes. Western blot and Real Time-PCR were used to detect the protein and mRNA expression levels of active Caspase-1 and Pyroptosis' s key protein Gasdermin-D( GSDMD). Results: Compared with NC group,the blood glucose and serum creatinine of DM group increased. HE and Masson staining showed some deposition in the renal tissue glomerular matrix and collagen fibrosis,swollen renal tubular epithelial cells,vacuolar degeneration,luminal dilatation and thickened basement membrane. The expression of protein and mRNA of active Caspase-1 and GSDMD was higher in DM group than that in NC group,and the difference was statistically significant( P < 0. 05). Conclusion: Caspase-1 activated GSDMD triggered pyroptosis may be induced in kidney tissues of DM.
