辛芍组方中灯盏细辛、赤芍单方对脑缺血再灌注损伤大鼠的作用及机制Protective Effects of Herba Erigerontis and Radix Paeoniae Rubra on Cerebral Ischemia Reperfusion Injury of Rats and Its Mechanism

摘要(Abstract):
目的:研究辛芍组方中灯盏细辛、赤芍单方对脑缺血再灌注损伤的作用及其机制。方法:将大鼠随机分为假手术组、模型组、阳性(灯盏花素)组、灯盏细辛组、赤芍组,术前7 d给予相应药物处理,模型组、阳性组、灯盏细辛组及赤芍组大鼠采用线栓法建立大鼠中动脉缺血再灌注损伤模型(MCAO),假手术组及模型组给予等量生理盐水;缺血2 h再灌注22 h后对大鼠进行神经行为学评分,观察脑梗死体积比,取血测定血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、一氧化氮(NO)、诱导型一氧化氮合酶(i NOS)、总一氧化氮合酶(TNOS)的含量; HE染色观察脑组织海马CA1区病理形态学变化,免疫组织化学法测定海马CA1区Bcl-2、Bax、Caspase-3蛋白的表达。结果:与模型组相比,两个单方均能不同程度改善大鼠神经行为学评分(P <0. 05),减小脑梗死体积(P <0. 05),血清SOD和GSH-PX活力增加(P <0. 05或P <0. 01)、MDA含量降低(P <0. 05),i NOS和TNOS活力降低(P <0. 05)、NO含量降低(P <0. 05),Bcl-2蛋白表达增加(P <0. 05或P <0. 01)、Bax和Caspase-3蛋白表达降低(P <0. 05或P <0. 01)。结论:灯盏细辛和赤芍单方可减轻大鼠脑缺血再灌注损伤,其机制可能与其减轻自由基损伤及提高Bcl-2蛋白表达有关。
Objective: To investigate the mechanism and protective effects of Herba Erigerontis and Radix Paeoniae Rubra on cerebral ischemia reperfusion injury. Methods: The rats were randomly divided into five groups: the sham operation group,the model group,Breviscapine group,Herba Erigerontis group and Radix Paeoniae Rubra group. After administration for 7 days,the intra-arterial ischemia reperfusion injury model(MCAO) was established. After ischemia for 2 h and reperfusion for 22 h,the rats were scored according to their neurobehavioral behavior; TTC staining was used to observe the volume ratio of cerebral infarction. Serum levels of MDA,SOD,GSH-PX,NO,i NOS and TNOS were examined. The hippocampal CA1 change of brain tissues was observed by HE staining. The expressions of Bcl-2,Bax and Caspase-3 protein in hippocampal CA1 region were determined by immunohistochemistry(IHC). Results: It was found that Herba Erigerontis and Radix Paeoniae Rubra could significantly reduce the symptoms of nervous function and the volume of cerebral infarction in rats with MACO(P < 0. 05). Compared with the model group,the rats in Herba Erigerontis and Radix Paeoniae Rubra group showd increased activities of SOD and GSH-PX(P < 0. 05 or P < 0. 01),and decreased activities of i NOS and TNOS(P < 0. 05),and that the contents of NO and MDA decreased in Herba Erigerontis and Radix Paeoniae Rubra group(P < 0. 05). In addition,Herba Erigerontis and Radix Paeoniae Rubra could upregulate the protein level of Bcl-2 and downregulate it of Bax and caspase-3 in MCAO rats(P < 0. 05 or P < 0. 01). Conclusion: Herba Erigerontis and Radix Paeoniae Rubra showed anti-cerebral ischemia reperfusion injury,which may be associated with the ability of anti-free radical damage and regulation of apoptosis-related proteins.
