股静脉置管注射垂体后叶素制作大鼠心肌缺血模型An Experimental Study on Establishment of Rat Myocardial Ischemia Model with Pituitrin

摘要(Abstract):
目的:探讨垂体后叶素(P it)制作大鼠心肌缺血模型的的最佳方法。方法:通过比较肌肉注射、股静脉、腹腔注射P it方法,股静脉置管注入不同剂量P it,结合心电监测及心肌肌钙蛋白等生化指标和病理切片,评估不同给药方法和股静脉置管注入不同剂量P it的造模效果。结果:股静脉滴注P it造成心肌缺血时间>60m in,与其它给药方式比较,P<0.01;动物模型的心肌缺血效应及不良反应事件与P it用量呈正相关。结论:股静脉置管滴注P it制作大鼠心肌缺血模型的方法可靠。目的:探讨垂体后叶素(Pit)制作大鼠心肌缺血模型的的最佳方法。方法:通过比较肌肉注射、股静脉、腹腔注射Pit方法,股静脉置管注入不同剂量Pit,结合心电监测及心肌肌钙蛋白等生化指标和病理切片,评估不同给药方法和股静脉置管注入不同剂量Pit的造模效果。结果:股静脉滴注Pit造成心肌缺血时间>60min,与其它给药方式比较,P<0.01;动物模型的心肌缺血效应及不良反应事件与Pit用量呈正相关。结论:股静脉置管滴注Pit制作大鼠心肌缺血模型的方法可靠。
Objective: To explore proper medication path and dose of pituitrin(Pit) for establisment of rat myocardial ischemia model,and to study characteristics of the model.Methods: Several medication methods including femoral vein injection,intraperitoneal injection,and intramuscular injection and doses were compared.The modelling effects were assessed with biochemistry indexes such as cardiac troponin(CTn),and superoxide dismutase(SOD),and pathological section observation combined with ECG monitoring.Results: Effect of myocardial ischemia and adverse reaction correlated positively with the doses of Pit used.Conclusions:(1) intravenous administration is more reliable and stable;(2) The proper dose should be 30 U/kg;(3) Pit can cause acute myocardial ischemia but not pathological change of myocardial infarction.
