糖通饮对2型糖尿病合并非酒精性脂肪肝大鼠肝组织的保护作用及机制
The protective effect of tangtongyin formula on type2diabetes mellitus complicated with rat non-alcoholic fatty liver disease and its mechanism

摘要(Abstract):

目的 研究糖通饮对2型糖尿病(T2DM)合并非酒精性脂肪肝模型大鼠肝组织的保护作用及可能机制。方法 44只SPF级SD雄性大鼠,随机抽取10只作为正常对照组予基础饲料喂养,其余大鼠予高脂高糖饲料喂养及低剂量链脲佐菌素(STZ)腹腔注射以建立T2DM合并非酒精性脂肪肝模型,并随机分为模型组、糖通饮(低、中、高)剂量组(相应剂量的糖通饮灌胃治疗8周);检测各组大鼠空腹血糖(FBG),测定血清甘油三酯(TG)、总胆固醇(TC)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、丙二醛(MDA)、超氧化物歧化酶(SOD)及肿瘤坏死因子-α(TNF-α)水平,采用HE染色法观察肝组织病理学变化,油红O染色法观察肝细胞脂质沉积情况。结果 与正常对照组比较,模型组大鼠肝组织脂质沉积增多,存在明显脂肪样变性,FBG及血清TG、TC、ALT、AST、MDA、SOD、TNF-α水平均明显升高(P<0.01);与模型组比较,各剂量组大鼠肝组织脂质沉积减少,脂肪样变性明显减轻,FBG及血清TG、TC、ALT、AST、MDA、SOD、TNF-α水平均有不同程度降低(P<0.05或P<0.01)。结论 糖通饮对T2DM合并非酒精性脂肪肝模型大鼠肝组织具有一定的保护作用,其机制可能与改善肝脏脂代谢紊乱、提高抗氧化能力、减轻炎症反应以及脂质过氧化损伤有关。
Objective To investigate the effect of Tangtongyin formula on the liver tissues of rats with type 2 diabetes mellitus(T2DM) complicated with non-alcoholic fatty liver disease(NAFLD)and its possible mechanism.Methods Forty-four male SD rats were randomly divided into normal control group(n= 10, fed with basal diet) and T2DM complicated with NAFLD model(n= 34, fed with high-fat & high-sugar diet and intraperitoneally injected with low-dose streptozotocin( STZ)).These rats with T2DM complicated with NAFLD were randomly divided into model group, low-dose,medium-dose, and high-dose groups based on Tangtongyin formula doses. Tangtongyin formula was given by gavage administration for 8 consecutive weeks. The levels of fasting blood glucose(FBG),serum triglyceride( TG), total cholesterol( TC), alanine aminotransferase( ALT), aspartate aminotransferase( AST), malondialdehyde( MDA), superoxide dismutation( SOD), and tumor necrosis factor-α(TNF-α) were examined. H&E staining was applied to observe pathological changes of liver tissues, and oil red O staining was performed to observe lipid accumulation.Results When compared with normal group, model group showed increased lipid deposition and significant steatosis in liver tissues and elevated levels of FBG and serum TG, TC, ALT, AST, MDA, SOD, and TNF-α(P< 0. 01). When compared with model group, lipid deposition and steatosis in liver tissues in each dose group were decreased, and the levels of FBG and serum TG, TC, ALT, AST, MDA, SOD, and TNF-α were reduced at different degrees(P< 0. 05 orP< 0. 01).Conclusion Tangtongyin formula has a certain protective effect on the liver tissues of rats with TDM complicated with NAFLD. Its mechanism may be related to improving lipid metabolism disorder, enhancing antioxidant capacity, and reducing inflammatory response and lipid peroxidation damage.
