抗菌肽Cecropin4与抗生素联合的体外抗菌效果

摘要(Abstract):

目的:探究抗菌肽Cecropin4(Cec4)分别与3种抗生素联合使用对鲍曼不动杆菌(AB)的体外抗菌效果。方法:采用微量肉汤稀释法测定抗菌肽Cec4、注射用头孢哌酮钠舒巴坦钠(SCF)、注射用亚胺培南西司他丁钠(IMP)及注射用硫酸多黏菌素B(PB)单用时对AB的最小抑菌浓度(MIC),运用棋盘法检测抗菌肽Cec4分别与这3种抗生素联用时对AB的抑菌效果,用部分抑菌浓度指数(FIC index)反映抗菌肽Cec4分别与这3种抗生素联用对AB的用药效果。结果:抗菌肽Cec4、SCF、IMP及PB对标准、多重耐药和泛耐药AB时菌株的MIC分别为4、16~64、16~64及0.625 mg/L;抗菌肽Cec4与PB联合作用于标准AB时、FIC index为0.50,抗菌肽Cec4分别与3种抗生素联合作用于多重耐药和泛耐药AB时,FIC index范围均在0.5~1.0。结论:抗菌肽Cec4与PB联合使用对标准AB表现为协同作用,抗菌肽Cec4分别与3种抗生素联合对多重耐药和泛耐药AB表现为相加作用。
Objective: To explore the antibacterial effect of antimicrobial peptide Cecropin4(Cec4) against Acinetobacter baumannii in combination with three antibiotics in vitro. Methods: The minimal inhibitory concentration(MIC) of the drug against Acinetobacter baumannii was determined by micro-broth dilution method, and the four drugs are antimicrobial peptide Cec4, Cefoperazone Sodium and Sulbactam Sodium for Injection(SCF), Imipenem and Cilastatin Sodium for Injection(IMP) and Polymyxin B(PB). The checkerboard method was used to detect the antibacterial effect of antimicrobial peptide Cec4 combined with the above three antibiotics on Acinetobacter baumannii. The fractional inhibitory concentration index(FIC index) was used to reflect the effect of medication between the antimicrobial peptide Cec4 and three antibiotics. Results: The MIC of the Cec4, SCF, IMP and PB against standard, multi-drug, and extensive-drug resistant Acinetobacter baumannii was 4 mg/L, 16~64 mg/L, 16~64 mg/L and 0.625 mg/L respectively. When the antibacterial peptide Cec4 was used in combination with PB for standard Acinetobacter baumannii, the FIC index was 0.50. The antibacterial peptide Cec4 was used in combination with three antibiotics for multi-drug resistance and extensive-drug resistance Acinetobacter baumannii, and the FIC index ranged from 0.5 to 1. Conclusion: The antibacterial peptide Cec4 combined with PB showed synergistic effect on the standard Acinetobacter baumannii, and cooperated with three antibiotics for multi-drug resistance and extensive-drug resistance of Acinetobacter baumannii.
