摘要(Abstract):
目的:探讨大鼠下颌持续功能前导后髁突软骨各部位骨形态发生蛋白-2(BMP-2)的表达。方法:40只35 d龄雄性SD大鼠均分为实验组和对照组,实验组大鼠24 h戴自制功能性矫治器引导下颌前伸,对照组不做处理;于实验后第3、7、14及28天时处死大鼠,采用免疫组化方法观察髁突软骨前、中、后各部位骨组织中BMP-2的表达。结果:随着观测时间点后移,对照组中髁突软骨前、中、后部的BMP-2阳性细胞数逐渐减少,实验组BMP-2阳性细胞数逐渐增加,差异无统计学意义(P>0.05);各时点同组大鼠髁突软骨后部BMP-2表达阳性细胞数高于中部和前部(P<0.05);从第7天起,实验组BMP-2表达阳性细胞数高于同时间点同部位对照组,差异有统计学意义(P<0.05、0.01)。结论:BMP-2可能参与了下颌骨髁状突骨改建过程;但对不同部位软骨的改建可能不同。
Objective: To observe the expression of bone morphogenetic protein 2( BMP-2) in the condylar cartilage of rats during mandibular advancement. Method: Forty 35-day-old male SpragueDawley( SD) rats were randomly divided into experimental group and control group. In experimental group the rats were treated with self-made functional appliance which caused mandibular protrusion while in control group the rats received no treatment. 5 rats in each group were sacrificed on the day 3,7,14 and 28 after experiment respectively. The expressions of BMP-2 in anterior,middle and posterior region of condyle cartilage were examined by immunohistochemical staining. Results: With the observation time extended,the number of BMP-2 expression positive cells in anterior,middle and posterior region of condyle cartilage in control group decreased gradually while the number of BMP-2 positive cells increased gradually,but the differences between two groups were not statistically significant( P >0. 05). At different time points,the number of BMP-2 expression positive cells in posterior region was higher than that of anterior or middle region in experimental group( P < 0. 05). From 7th day,at the same time point,the number of BMP-2 expression positive cells in experiment group was significantly higher than that of control group( P < 0. 05). Conclusions: BMP-2 may participate in the remodeling activities of condylar cartilage. Its effect on mandibular cartilage remodeling may be different in different region of condyle cartilage.
