二氢麦角毒对MPTP帕金森病模型小鼠的作用A Study on Effect of Dihydroergotoxine on Parkinsonism Model Mice

摘要(Abstract):
目的:观察二氢麦角毒对1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)帕金森病模型小鼠行为学、脑超氧化物歧化酶(SOD)活性及纹状体多巴胺及其代谢产物含量的影响。方法:小鼠腹腔注射MPTP 30 mg/kg,2次/d,连续5 d致帕金森模型,用YSD-4G药理生理实验多用仪测定小鼠自发活动记数,用高效液相色谱(HPLC)法测定纹状体内多巴胺及其代谢产物含量,酶法测脑SOD活性。结果:二氢麦角毒能明显增加MPTP帕金森病模型小鼠的自发活动、脑SOD活性和纹状体内多巴胺及其代谢产物含量。结论:二氢麦角毒能改善MPTP帕金森病模型小鼠的自发活动减少的症状,其机制可能与增加脑SOD活性及纹状体内多巴胺含量有关。
Objective: To observe the effects of dihydroergotoxine on parkinsonism model mice including their behavior, activity of brain SOD, and the contents of dopamine and derivatives in striate body. Methods: Parkinsonism model mice were established by giving mice with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) (ip) at a dosage of 30mg/kg, twice a day for 5 days. The locomotor activity was measured on the 7th and 14th days afterwards. The contents of dopamine, dihydroxyphenylacetic acid and homovanillic acid in striatum were measured with LPHC, and SOD activity in brain tissue was measured with enzyme method. Results: The locomotor activity, and the contents of dopamine, dihydroxyphenylacetic acid, homovanillic acid and SOD increased significantly in dihydroergotoxine groups. Conclusion: Dihydroergotoxine can remarkably enhance the locomotor activity of the MPTP-treated mice, which might relate with increase of dopamine, dihydroxyphenylacetic acid, homovanillic acid and SOD in brain tissue.
