低频电刺激双侧丘脑底核对癫痫模型大鼠脑内谷氨酸表达及海马细胞单位放电的影响
Influence of Low-frequency Electrical Stimulation of Bilateral Subthalamic Nucleus on the Glutamate Expression in Brain Tissues and Unit Discharges of Hippocampal Neurons of Epileptic Model Rats

摘要(Abstract):

目的:观察低频电刺激(LFS)双侧丘脑底核(STN)对杏仁核电点燃癫痫模型大鼠脑内谷氨酸(Glu)表达及海马细胞单位放电(UCD)的影响。方法:健康雄性Wistar大鼠30只随机分为对照组、癫痫模型组和LFS组,各组大鼠杏仁核均埋置刺激电极,对照组不刺激杏仁核,癫痫模型组及LFS组均电刺激制作癫痫模型。LFS组在癫痫点燃模型成功后予LFS双侧STN,观察各组大鼠癫痫样行为学改变,并记录海马UCD及脑组织Glu阳性神经元的表达。结果:癫痫模型组海马UCD频率较对照组增多(P<0.05),LFS组海马UCD频率比模型组减少(P<0.05);癫痫模型组脑组织中Glu表达较对照组增高(P<0.05),LFS组脑组织中Glu的表达较癫痫模型组降低(P<0.05)。结论:LFS双侧STN可抑制杏仁核电点燃癫痫模型大鼠脑内海马UCD频率及Glu阳性神经元的表达。
Objective: To observe the influence of low frequency electrical stimulation( LFS) of bilateral subthalamic nucleus( STN) on glutamate( Glu) expression in brain tissues and unit discharges of hippocampal neurons( UCD) of amygdaloid kindling epilepsy rat model. Methods: Thirty healthy male Wistar rats were randomly divided into control group,epileptic model group and LFS group. Stimulating electrodes were buried in amygdale of rats in the three groups,and amygdale of rats in model group and LFS group was stimulated to establish epileptic model,that of control group was not stimulated. After modeling,LFS group was given LFS to bilateral STN. Epileptics behaviors of rats in each group were observed and UCD were recorded in each group and the expression of Glu was determined. Results: UCD frequency in epileptic model group was more than that in control group,LFS group was less than model group( P ﹤ 0. 05). The expression level of Glu in epileptic model group was higher than that in control group,LFS group was lower than model group( P ﹤ 0. 05). Conclusions: LFS may significantly inhibit the expression of glutamate and decrease the UCD frequency in amygdaloid kindling rat model.
