HPLC同时测定蜘蛛香中9种指标成分的含量Simultaneous Determination of Nine Components in Valeriana jatamansi Jones by HPLC

摘要(Abstract):
目的:建立高效液相色谱法(HPLC)同时测定蜘蛛香中新绿原酸、绿原酸、咖啡酸、异绿原酸B、橙皮苷、异绿原酸A、异绿原酸C、乙酰缬草三酯及缬草三酯含量的方法。方法:采用DiamonSil~? C_(18)色谱柱,乙腈(A)-0.1%甲酸水溶液(B)为流动相梯度洗脱,检测波长为327 nm(0~33 min)、256 nm(33~90 min),流速为1.0 mL/min,柱温为30℃。结果:新绿原酸在0.75~120.00 mg/L(r=1.000 0)、绿原酸在5.23~418.00 mg/L(r=1.000 0)、咖啡酸在0.44~44.00 mg/L(r=1.000 0)、异绿原酸B在2.1~210.00 mg/L(r=1.000 0)、橙皮苷在2.20~220.00 mg/L(r=1.000 0)、异绿原酸A在2.065~330.000 mg/L(r=0.999 9)、异绿原酸C在1.88~150.00 mg/L(r=1.000 0)、乙酰缬草三酯在2.50~200.00 mg/L(r=1.000 0)、缬草三酯在10.00~800.00 mg/L(r=1.000 0)的进样浓度范围内线性关系良好,加样回收率(n=6)为101.92%~107.51%,RSD<3%;25批蜘蛛香样品中9种成分含量差异较大,其中缬草三酯、橙皮苷、异绿原酸A及绿原酸等4个成分的含量较高。结论:HPLC可同时测定蜘蛛香中9种成分含量。
Objective: To establish an HPLC method for the simultaneous determination of nine constituents in Valeriana jatamansi,including neochlorogenic acid, chlorogenic acid, caffeic acid, hesperidin, isochlorogenic acid B, isochlorogenic acid A, isochlorogenic acid C, acevaltrate and valepotriate. Methods: The analysis of this drug was performed on a DiamonSil~? C_(18) column, with acetonitrile(A)-0.1% formic acid(B) solution as the mobile phase for gradient elution, the detection wavelength at 327 nm(0~33 min) and 256 nm(33~90 min), the flow rate at 1.0 mL/min, and the column temperature at 30 ℃. Results: Good linear relationships of neochlorogenic acid, chlorogenic acid, caffeic acid, hesperidin, isochlorogenic acid B, isochlorogenic acid A, isochlorogenic acid C, acevaltrate and valepotriate was showed within the ranges of 0.75~120.00 mg/L(r=1.000 0),5.23~418.00 mg/L(r=1.000 0),0.44~44.00 mg/L(r=1.000 0),2.10~210.00 mg/L(r=1.000 0),2.20~220.00 mg/L(r=1.000 0),2.065~330.000 mg/L(r=0.999 9),1.88~150.00 mg/L(r=1.000 0),2.50~200.00 mg/L(r=1.000 0),10.00~800.00 mg/L(r=1.000 0) respectively; the average recoverie rates(n=6) were between 101.92%~107.51% and each RSD was under 3%; the contents of nine components in 25 batches of Valeriana Jatamansi Jones were different, and the contents of valepotriate, hesperidin, isochlorogenic acid A and chlorogenic acid were the highest. Conclusion: HPLC can be used for simultaneous determination of 9 components in Valeriana Jatamansi.
