氧化苦参碱微球体外累积释放百分率的影响因素

摘要(Abstract):

目的:考察改善氧化苦参碱(OMT)聚乳酸聚乙醇酸(PLGA)微球(MS)24 h累积释放百分率的制备工艺。方法:采用乳化溶剂挥发法(W/O/W)制备MS,保证载药量变化不大的情况下,以24 h累积释放百分率为指标考察聚乙烯酸(PVA)浓度、吐温浓度、搅拌速度、Na Cl浓度、PLGA浓度、外水相体积对MS体外释放的影响。结果:最优处方为PVA浓度为1%,Na Cl浓度为2%,PLGA浓度为125 g/L,外水相体积为100 m L,OMTMS载药量和包封率分别为28.95%、93.27%,24 h累积释放百分率为8.11%。结论:在该条件下制备OMT-MS24 h突释率得到改善。
Objective: To explore preparation technics for improvement of 24 h cumulative release percentage of oxymatrine( OMT) poly( 1actic acid-co-glycolie acid)( PLGA) microspheres( MS).Methods: By the Emulsified solvent evaporation( W/O/W),the effects of the concentration of PVA,Tween,Na Cl,PLGA and the stirring speed,external water phase volume on cumulative release percentage of OMT MS were investigated in the case of small changes in drug loading. Results: The optimal prescription: the concentrations of PVA,Na Cl,and PLGA were 1 %,2 %; and 125 g/L respectively; the volume of external water was 100 m L; the drug-loading rate and encapsulation efficiency of OMT-MS were 28. 95% and 93. 27% respectively,and the 24 h cumulative release percentage was8. 11%. Conclusion: The 24 h cumulative release percentage of microspheres could be improved when OMT-MS prepared under the mentioned condition.
