青藤碱对胰腺癌细胞系SW1990增殖及凋亡的影响Effect of Sinomenine on Pancreatic Cancer Cell SW1990 Proliferation and Apoptosis

摘要(Abstract):
目的:探讨青藤碱(SIN)对胰腺癌细胞增殖及凋亡的影响。方法:胰腺癌SW1900细胞分为实验组和对照组,实验组设置SIN浓度梯度0.125、0.25、0.5、1.0及2.0 mmol/L组,对照组不作处理;CCK-8方法检测SIN对胰腺癌细胞SW1990增殖的影响,平板克隆实验检测SIN对胰腺癌SW1990细胞集落形成的影响,流式细胞术检测SIN对胰腺癌SW1990细胞凋亡的影响,Western blot检测SIN对胰腺癌SW1990细胞中PARP蛋白表达水平的影响。结果:SIN作用于SW1990细胞后,细胞增殖率随着SIN剂量增加而降低(P<0.05),细胞集落形成数量逐渐减少(P<0.05),细胞凋亡率逐渐增加(P<0.05);相同浓度SIN时,随着作用时间延长,其对SW1990细胞增殖的抑制作用逐渐增强(P<0.05);Western blot结果显示胰腺癌SW1990细胞PARP蛋白表达水平逐渐随着SIN剂量增加而减少(P<0.05)。结论:青藤碱能有效抑制胰腺癌SW1990细胞的增殖,其机制可能与诱导SW1990细胞凋亡有关。
Objective: To investigate the effect of Sinomenine on proliferation and apoptosis of pancreatic cancer cell SW1990. Methods: Human pancreatic cancer cell line SW1990 cells were divided into experiment group and control group; experiment group was further divided into 0. 125,0. 25,0. 5,1. 0 and 2. 0 mmol/L group by concentrations of SIN,while control group receive none treatment.CCK-8,plate cloning experiment,flow cytometry and Western blot were adopted to test the effect of SIN on pancreatic cancer cell SW1990 proliferation activity,colony formation,apoptosis,and protein expression level of PARP respectively. Results: Experiment showed that cell proliferation rate decreased as the SIN dosage increased( P < 0. 05),colony formation decreased gradually( P < 0. 05),apoptosis rate increased gradually( P < 0. 05); with the same concentration level,as the time prolonged,the inhibition effect on SW1990 cell proliferation enhanced( P < 0. 05); Western blot showed that protein expression level of PARP gradually increased as the SIN dosage added( P < 0. 05). Conclusion: These results suggest that Sinomenine could inhibit proliferation and induce apoptosis in human pancreatic cancer cell SW1990.
