下调接头蛋白CrkII对喉癌Hep-2细胞生物学行为的影响Effects of Down-regulated Junction Protein CrkII on Biological Behaviour of Laryngeal Carcinoma Hep-2 Cells

摘要(Abstract):
目的:探讨下调接头蛋白Crk II对喉癌Hep-2细胞增殖、迁移和侵袭能力的影响。方法:采用RNA干扰法敲低Crk II的表达,以脂质体转染法将质粒导入Hep-2细胞中,通过real-time PCR和Western blot分别检测各组细胞中Crk II mRNA和蛋白的水平,MTT、细胞划痕及Transwell侵袭实验检测转染后细胞增殖、迁移和侵袭能力的改变。结果:质粒转染后能有效的抑制Hep细胞Crk IImRNA和蛋白表达水平(P<0.01),Hep-2细胞增殖、迁移和侵袭能力明显降低(P<0.01)。结论:Crk II基因沉默可明显抑制Hep-2细胞的增殖、迁移与侵袭,接头蛋白Crk II可促进喉癌Hep-2细胞的恶性生物学行为。
Objective:To investigate the effect of down-regulated junction protein CrkII on the proliferation,migration and invasion of laryngeal carcinoma Hep-2 cells.Methods:The expression of CrkII was reduced by RNA interference,and the plasmid was transfected into Hep-2 cells by liposome transfection.The levels of CrkII mRNA and protein were detected by real-time PCR and Western blot,respectively.MTT,Cell scratch and Transwell invasion assay were used to detect the changes of cell proliferation,migration and invasion ability after transfection.Results:Plasmid transfection could effectively inhibit the expression of CrkII mRNA and protein in Hep cells(P<0.01).Proliferation,migration and invasion of Hep-2 cells decreased significantly(P<0.01).Conclusion:CrkII gene silencing can significantly inhibit the proliferation,migration and invasion of Hep-2 cells.CrkII can promote the malignant biological behavior of Hep-2 cells in laryngeal carcinoma.
