瑞香狼毒素亚慢性染毒对大鼠胃组织的影响Effect of subchronic exposure to Stellera chamaejasme toxin on the stomach of rats

摘要(Abstract):
目的探讨瑞香狼毒素亚慢染毒对大鼠胃组织的损害作用及可能机制。方法 50只SPF级SD大鼠,雌雄各半,随机分为对照组(去离子水灌胃90 d)、(低、中、高)剂量组(分别以31.25、125、500 mg/kg瑞香狼毒素水溶液灌胃90 d)以及恢复组(以500 mg/kg染毒剂量灌胃90 d后,自然恢复7d);每周观测大鼠体质量,采用胶乳增强免疫比浊法检测血清胃蛋白酶原(PG)Ⅰ和Ⅱ的含量,采用石蜡切片观察胃组织的病理组织学变化,免疫印迹法测胃组织匀浆缝隙连接蛋白43(CX43)的蛋白表达。结果与对照组相比,中剂量组大鼠血清PGI含量、胃组织匀浆中CX43蛋白表达量降低(P<0.05),高剂量组大鼠体质量、血清PGI和PGII含量、胃组织匀浆中CX43蛋白表达量降低(P<0.05);高剂量组PGI含量、CX43蛋白表达量低于低剂量组(P<0.05);高剂量组大鼠的胃黏膜上皮缺损,有不典型增生,胃腺萎缩和纤维化;与高剂量组相比,恢复组大鼠血清PGI和PGII含量、胃组织匀浆中CX43蛋白表达量以及胃组织病理观察基本无变化。结论瑞香狼毒素较长时期暴露可引起胃组织损伤,这可能与CX43的表达受抑制有关。
Objective To investigate effect of subchronic exposure to the Stellera chamaejasme toxin on rats' stomach tissue and possible mechanism.Methods Fifty SPF SD rats in equal gender parity were randomly divided into control group(intragastric administration of deionized water for 90 days),the low,medium,and high-dose group(intragastric administration of S.chamaejasme toxin solution31.25,125,and 500 mg/kg for 90 days),and recovery group(90 days of intragastric administration of 500 mg/kg toxin solution,then recovered naturally for 7 days).The rats were weighed every week.The contents of PGⅠ and PG Ⅱ in serum were detected by latex enhanced immunoturbidimetry.The histopathological changes of the stomach tissues were observed by the paraffin section,and the protein expression of gastric homogenate gap connexin 43 was measured by immunoblotting.Results Compared with the control group,serum PGI content and expression of CX43 in gastric tissue homogenate of medium group were decreased(P <0.05);body weight,content of serum PGI and PGII,expression of CX43 in gastric tissue homogenate of high dose group were decreased(P <0.05),and PGI content and expression of CX43 of high dose group were lower than that of low dose group(P <0.05).Pathology study showed that gastric mucosal epithelial defect with atypical hyperplasia,gastric gland atrophy,and fibrosis in the high-dose group;compared with the high-dose group,there was no changes in serum PGⅠ and PGⅡ content,CX43 protein expression and pathological changes in the recovery group.Conclusion Long-term exposure to S.chamaejasme toxin may cause gastric tissue damage,which may be related to the inhibition of CX43 expression.
