贵阳地区常见嗜尸性蝇类丽蝇科DNA条形码的构建Establishment of DNA Barcode of Common Calliphoridae Sarcosaprophagous Fly in Guiyang City

摘要(Abstract):
目的:构建贵阳地区常见嗜尸性蝇类丽蝇科的系统发育树。方法:对家兔尸体上采集到的嗜尸性蝇类丽蝇科的卵、幼虫及成虫提取DNA,扩增COI基因条形码区的序列,采用CLC Sequence View、MEGA软件进行序列分析,构建系统发育树。结果:共采集到丽蝇科5属9种嗜尸性蝇类,经序列测定确定各样本的形态学鉴定与分子鉴定是符合的,并以此序列构建出贵阳地区常见嗜尸性蝇类丽蝇科的系统发育树。结论:可以利用COⅠ基因条形码区序列构建的系统发育树进行贵阳地区嗜尸性蝇类的种属鉴定。
Objective: To establish the phylogenetic tree of common calliphoridae sarcosaprophagous fly in Guiyang city,and so as to provide basis for identification of eggs and larvae of calliphoridae sarcosaprophagous fly in Guiyang city.Methods: DNA of eggs,larvae and adults of sarcosaprophagous fly(calliphoridae) was extracted from rabbit corpse.Sequence of barcode region of cytochrome oxidase I(COI) gene was amplified by PCR.Softwares such as CLC sequence view and MEGA were employed to analyze the sequence.The phylogenetic tree of sarcosaprophagous fly was established.Results: A total of 5 genus and 9 species calliphoridae sarcosaprophagous fly were collected.After identifying the sequences and removing primers and same sequences,the phylogenetic tree of calliphoridae sarcosaprophagous fly in Guiyang city was established.Conclusions: The phylogenetic tree of calliphoridae sarcosaprophagous fly in Guiyang city is established based on sequence of barcode region of COI gene,which provides the basis for identification of calliphoridae sarcosaprophagous fly in Guiyang.
