慢性染砷对大鼠大脑皮层组织细胞DNA损伤及细胞凋亡的影响

摘要(Abstract):

目的:通过单细胞凝胶电泳(SCGE)和免疫组织化学的方法探讨慢性染砷对大鼠大脑皮层组织细胞的影响及机制。方法:60只3周龄Wistar大鼠随机分为低剂量染砷组(A1)、高剂量染砷组(A2)和对照组(C),分别自由饮用10.0 mg/L、40.0 mg/L的三氧化二砷水溶液和自来水,喂养6个月后处死大鼠,取新鲜的大脑皮层进行SCGE实验,检测各组大脑皮层凋亡蛋白Bax的表达。结果:SCGE实验提示,染砷组大脑皮层细胞DNA损伤明显高于对照组,P<0.01。凋亡蛋白Bax在3组均有表达,随着染毒剂量的增加Bax表达增强,A2组与C组比较P<0.01。结论:慢性染砷可引起大鼠大脑皮层组织细胞DNA损伤和诱导细胞凋亡。
Objective: To investigate the effects of chronic arsenic poisoning on the cerebral cortex of rats and the possible mechanism.Methods: Sixty-three-weekold Wistar rats were randomly divided into three groups as following: low dose arsenic poisoning group(A1),high dose arsenic poisoning group(A2),and control group(C).Rats in group A1 and group A2 were fed with water which was dissolved with arsenic trioxide at the concentrations of 10.0mg/L and 40.0mg/L respectively.Rats of control group were fed with normal tap water.In 6 months later,the rats were sacrificed and single suspensions of cell were prepared with fresh cerebral cortex tissues and the suspensions were then subjected to single cell gel electrophoresis(SCGE).Expressions of cerebral cortex apoptosis protein Bax were assayed by using immunohistochemistry methods.Results: DNA injuries of groups A1 and A2 were much more serious than that in control group(P<0.01).Bax expression occurred in all of the three groups,and the expression extent significantly increased following the increase of arsenic trioxide intake.Conclusion: Chronic arsenic poisoning can result in DNA injuries and apoptosis of cerebral cortex in rats.
