臭氧治疗对家兔骨性关节炎病变及NO含量的影响The Impact of Ozone Treatment to Rabbit Osteoarthritis and Nitrogen Monoxidum Contents

摘要(Abstract):
目的:探讨臭氧治疗对家兔骨性关节炎(OA)关节冲洗液中NO含量的影响。方法:40只新西兰兔随机分为5组,A组为正常对照组,B组为模型对照组,C、D、E组分别为臭氧(20、40、60 mg/L)治疗组;用Vand-m an法造模型成功后,将不同浓度的臭氧注射入C、D、E组兔膝关节腔内,每3天1次,连续2次;检测关节冲洗液中NO的含量,观察关节软骨的病理变化并进行病理评分。结果:B组软骨呈典型的OA改变,C、D组关节软骨可见不同程度修复,病理评分与B组相比有显著性差异(P<0.01或P<0.05);E组未见明显修复,病理评分与B组无显著差异;与A组相比,B组关节冲洗液中NO含量显著增高(P<0.01);与B组相比,C、D治疗组NO含量显著降低(P<0.01),E组NO显著增高(P<0.01)。结论:20~40 mg/L浓度的臭氧可促进实验性OA关节软骨修复,其机制可能与抑制NO生成有关,60 mg/L浓度的臭氧无软骨修复作用。
Objective: To explore the impact of ozone treatment to rabbit osteoarthritis(OA) and nitrogen monoxidum(NO) contents in joint flushing fluid(JFF).Methods: Forty New Zealand rabbits were randomly divided into five groups: normal control group(group A),model control group(group B),and treatment groups C,D,and E which were treated with ozone at doses of 20 mg/L,40 mg/L,and 60 mg/L respectively.Models were established with Vandman method successfully,and different doses of ozone were injected into rabbit knee joint cavity of groups C,D,and E twice a week.NO contents in JFF and articular cartilage variation were determined and pathological changes in arthrodial cartilage were observed and analyzed.Results: Typical OA changes in cartilage were found in group B,while different degrees of repairing on articular cartilage could be seen in groups C and D of which the pathological scores differed from that of group B significantly(P<0.01,P<0.05),No obvious repairing was found in group E,of which the pathological score did not differ from that of group B.NO content in JFF was significantly higher in group B than in group A(P<0.01);NO contents in JFF of groups C and D were obviously lower than that in group B(P<0.01),while in group E,NO content significantly increased(P<0.01).Conclusions: Ozone with concentrations of 20~40 mg/L can promote articular cartilage repair in experimental OA,and its mechanism may relate to inhibiting NO generation.Ozone of 60 mg/L has no repairing effect to cartilage.
