登革热病毒Ⅱ型临床株感染小鼠TH1/TH2类细胞因子的产生A Research on TH1/TH2 Cytokines in Balb/c Mice Infected with Different Clinic Strains of Type 2 Dengue Virus

摘要(Abstract):
目的:观察登革病毒2型临床分离株B株(DEN2B)感染Balb/c小鼠后,其血浆中γ-干扰素(IFN-γ)、白细胞介素2,4,5,6,10(IL-2,IL-4,IL-5,IL-6,IL-10)、β-转化生长因子(TGF-β)的动态变化,并探讨DEN2B株感染后小鼠Ⅰ型辅助性T细胞(TH1)、Ⅱ型辅助性T细胞(TH2)应答机制及其相关关系。方法:1.用不同剂量DEN2B株经皮下多点注射感染Balb/c小鼠,建立动物模型;2.双抗体夹心ELISA法检测不同剂量DEN2B株感染后各组动物血浆中(体内)IFN-γ,IL-2,IL-4,IL-5,IL-6,IL-10,TGF-β的产生动态。结果:1.DEN2B株感染动物后,各剂量组动物血浆中均检出特异性抗DEN2IgM,IgG类抗体,出现病毒血症及不同程度临床表现,建立了DEN2B株感染Balb/c小鼠动物模型;2.DEN2B株感染动物后,其血浆IFN-γ,IL-2,IL-4,IL-5,IL-6,IL-10,TGF-β水平在感染后不同时期均出现不同程度增加,各种细胞因子水平变化情况与正常组差别有统计学意义(P<0.05)。各种细胞因子产生动态有差异。结论:IFN-γ,IL-2,IL-4,IL-5,IL-6,IL-10,TGF-β等细胞因子与登革病毒感染所致疾病病情关系密切,DEN2B株再次感染阶段以TH2应答为主。
Objective: To study the mechanism of immune response of TH1/TH2 in Balb/c mouse infected with clinic strain (B strain) of DEN2 virus and their relations. Methods: 1) The Balb/c mouse infection model was established by multisubcutaneous injection with various doses of DEN2 strain B. 2) Mouse plasma samples collected from different experiment groups at various time points after infection were tested for IL-4,IFN-γ,IL-2,IL-5,IL-6,IL-10 and TGF-β levels with sandwich ELISA. Results: Balb/c mouse infection model was successfully established. The mice presented clinical manifestations including fever, and homologous pathologic changes. Viremia and special antibodies to strain B of DEN2 were detected in the samples after infection; 2) Increased levels of cytokines such as IL-4, IFN-γ, IL-2, IL-5, IL-6, IL-10 and TGF-β were observed in the serum of infected mice. The dynamic levels of IFN-γ and IL-4 produced by infected animals were associated with the doses of DEN2. Levels of both TH1 and TH2 cytokines including IFN-γ, and IL-4 increased after the first infection. And all of the cytokines increased after reinfection. But TH2 cytokines were higher than TH1 cytokines in the reinfection phase. Conclusions: The cytokines such as IFN-γ, IL-2, IL-4, IL-5, IL-6, IL-10, and TGF-β may play important roles in the infection caused by DEN virus. Responses of both TH1 and TH2 can be induced by DEN2 strain B in the first infection phase, while TH2 response is more predominant than TH1 response in the second infection phase.
