地诺帕明对·OH所致麻醉大鼠异常血流动力学的保护作用Protective Effects of Denopamine on the Abnormal Hemodynamics Induced by·OH in Anaesthetized Rats

摘要(Abstract):
静注羟自由基(·OH)生成液0.5m·100g~(-1),使麻醉大鼠心功能明显受损,表现为左室收缩期峰值压(LVSP)及其与心率的乘积(LVSP×HR)、平均动脉压(MAP)、左室等容收缩期最大上升和下降速率(+dP/dtmax和-dP/dtmax明显降低,而左室舒张末期压(LVEDP)显著升高,在给·OH前5min静注地诺帕明(Deno)18μg·100g~(-1),再以3μg·100g~(-1)·min~(-1)于给·OH后立即静脉输注,可不同程度地改善上述异常血流动力学指标,基中对LVEDP改善尤为显著。Deno还明显对抗·OH所致血浆丙二醛含量的升高。
The effects of denopamine (Deno) on the abnormal hemodynamics induced by exogenous hydroxyl free radical (·OH) were investigated in anaesthetized rats. Intravenous administration of ·OH-generating solution ( H2O2 1 mmol with equimolar CuCl2 and vitamin C) 0. 5ml·100g1 significantly decreased LVSP,LVSP×HR, MAP, +dP/dtmax and -dP/dtmax, and increased LVS in anaesthetized rats. These effects maintained for the whole surveying period (1h). Preadministration (iv, 5 min before·OH) of Deno 18 μg·100-1 followed by intravenous infusion of 3μg.100g-1.·min-1 after administration of·OH significantly improved these abnormal hemodynamic indices induced by ·OH except EVSP which tended to be improved by Deno, the improvement of LVEDP was specifically significant. Ferthermore, the heightened serum malondialdehyde (MDA) content induced by ·OH was also decreased by Deno. The clinical significance was discussed.
