双偶合实验中副斑观测对区分难辨氨基酸的价值Study of By-Products Produced by Double Coupling of Amino Acids Using DABITC/PITC

摘要(Abstract):
用10种标准氨基酸进行模拟DABITC/PITC双偶合实验,结果表明:氨基酸可产生偶合副产物,副色斑的层析行为有助于各氨基酸的鉴别,同时讨论了影响副斑产生的因素。
The experiments of double coupling of 10 standard amino acids showed that some blue or/and red secondary spots can be obtained apart from the red spots attributed to the DABTH derivatives of the amino acids on polyamide thinlayer chromatography. By-products usually move with lower speed in either chromatographic dimension. Different colors and chromatographic properties of by-products appeared to be associated with parent amino acids. Characteristic byproducts are helpful in the correct identification of amino acids. Consant chromatcgraphic positions varying with the nature of the parent amin acid excluded the influences of reagents and uncleaved peptide derivatives. Neither were the secondary spots affected by the presence or absence of PITC in coupling step. Cleaving with TFA is disadvantageous to appearence of blue spot.
