神经肽Y在生后发育期间家兔直肠的表达及意义Expression of Neuropeptide Y in Rabbit Rectum during the Postnatal Development Period and Its Significance

摘要(Abstract):
目的:探讨生后发育期间家兔直肠内神经肽Y(NPY)的表达及分布特点。方法:取生后5、15、25、35、60、90 d及成年组家兔70只,取直肠组织,制成连续切片,用SABC法和图像分析法对NPY免疫反应阳性细胞的定位和表达进行研究。结果:NPY阳性细胞于生后5 d可见于家兔直肠黏膜,数量少,染色浅,多分布于上皮;35 d,NPY阳性细胞数量明显增多(P<0.05),60 d达到顶峰,90 d则减少至成人水平(P<0.05);主要分布于肠腺上皮。图像分析显示平均灰度值随家兔的生长发育不断降低(P<0.05)。结论:NPY在直肠的表达随家兔的生长发育显著增多,推测NPY参与调节生后发育期间家兔直肠消化功能的成熟过程。
Objective:To investigate the expression and distribution features of neuropeptide Y(NPY)in rabbit rectum during the postnatal development period.Methods: Rectums of 60 rabbits respectively aged 5,15,25,35,60,and 90 days,and 10 adult rabbits were taken to prepare serial sections.The localization and expression of NPY positive cells were detected with immunohistochemical SABC staining method and image analysis.Results: A few NPY positive cells scattered in epithelium mucosa were initially found in 5 days after birth.The cell number increased significantly on the 35th day(P<0.05),and reached the peak on the 60th day(P<0.05),but decreased afterwards and reached the adult level in 90 days(P<0.05).The positive cells mainly distributed in the epithelium of intestinal glands.Image analysis showed that the mean grey values of these cells decreased gradually(P<0.05)along with the growth of rabbits.Conclusions: The expression of NPY in rectum increases obviously with the early postnatal development of baby rabbit,which suggests NPY may participate in the regulation of digestive function maturation during the period.
