微血管密度在口腔癌前病变及口腔鳞癌中的意义A Study on the Role of Angiogenesis in Human Oral Precancerous Lesion and Oral Squamous-cell Carcinoma

摘要(Abstract):
目的:通过对口腔不同病变组织中微血管的定量分析,探讨血管生成在口腔癌前病变和癌变过程中的变化规律及其意义。方法:应用免疫组织化学二步法,以CD31为标记物对73例口腔良、恶性病变组织中的血管内皮细胞进行免疫组织化学染色,人工计数微血管密度(MVD)并进行定量分析。结果:口腔鳞癌和癌前病变组织中的MVD明显高于正常口腔黏膜(P<0.01),鳞癌组织MVD与肿瘤的分化、浸润和转移相关(P<0.01)。结论:血管生成与口腔癌前病变及癌变密切相关,癌前病变中的血管生成可反映癌前病变的恶性倾向。
Objective: To investigate the role of angiogenesis in the process of cancerating from oral precancerous lesion to oral carcinoma using quantitative analysis of microvessel density(MVD) in different oral tissue.Methods: Immunohistochemistry PV-9000 method was adopted,and 73 benign and malignant oral specimens were checked by immunohistochemical stainning with CD31 as a sensitive and specific marker for vascular endothelium.MVD were determined by manual count.Results: MVD was significantly higher in oral squamous-cell carcinoma(OSCC) and oral precancerous lesion than in normal oral mucosa(P<0.01). The MVD in OSCC was related to tumor differentiation,infiltration,and metastasis(P<0.01).Conclusions: Angiogenesis was closely related to malignant change of oral precancerous lesion.Presence of neovascularization in oral precancerous lesion might be an indicator of early malignant change.
