定心方及丹参酮ⅡA对心肌缺血-再灌注损伤大鼠白细胞介素-6的影响

摘要(Abstract):

目的:观察定心方及丹参酮ⅡA对心肌缺血-再灌注损伤大鼠血清白细胞介素-6(IL-6)含量的影响,探讨其对心肌的保护机制。方法:32只健康雄性Wistar大鼠,随机分为假手术组、模型组、定心方给药组、丹参酮ⅡA给药组,用冠脉结扎及再松开的方法制备心肌缺血-再灌注损伤动物模型;记录Ⅱ导联心电图,对心律失常进行评分;用HE染色观察心肌病理形态学变化,ELISA法检测血清IL-6含量。结果:定心方及丹参酮ⅡA干预后,心肌缺血-再灌注模型大鼠心律失常明显减少,心肌组织病理改变减轻,血清中IL-6表达显著降低,P<0.01。结论:定心方丹及参酮ⅡA可减少炎症因子IL-6大量释放,减轻炎症反应,改善受损心肌病理结构,发挥心肌保护作用。
Objective:To investigate the effects of Dingin recipe(DXR) and tanshinone ⅡA(Tan) on serum interleukin-6(IL-6) content in rat model of myocardial ischemia/reperfusion injury(MI/RI),and explore their mechanism of myocardial protection.Methods: Thirty-two male Wistar rats were randomly divided into four groups(8 in each):sham operation group(group SH),model group,group DXR and group Tan.MI/RI model was established by ligating left anterior descending branch of coronary artery.During the process,electrocardiogram was taking continuously to observe arrhythmias,and the serum level of IL-6 was measured by ELISA.At the same time,HE staining was performed to detect the pathological change in myocardium.Results: Serum IL-6 contents were lower and MI/RI was lightened significantly in groups DXR and Tan.Conclusions: Dingxin recipe and TanshinoneⅡA can decrease expression of IL-6 and pathological change of myocardium and reduce arrhythmia and inflammatory response after MI/RI in rats.These actions may contribute to the protective effect on the ischemic myocardial cells.
