脑通复方制剂对血管性痴呆大鼠治疗作用的探讨

摘要(Abstract):

目的:研究脑通复方制剂对血管性痴呆大鼠脑组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量的影响,探讨中药对血管性痴呆(VaD)大鼠空间学习记忆减退的防治作用。方法:采用改良Pulsinelli四血管阻断(4-VO)法制造VaD模型,设脑通复方制剂组、模型组、假手术组、盐酸多奈呱齐(安力申)组,分别测定干预后各组大鼠大脑海马组织中的SOD活性、MDA含量。结果:灌胃30 d后,与模型组相比,脑通复方制剂可改善VaD大鼠学习记忆能力,显著提高大鼠海马组织中SOD活性、明显降低MDA含量。结论:脑通复方制剂对血管性痴呆大鼠空间学习记忆减退有明显防治作用,其机制可能与其提高VaD大鼠脑组织中SOD活性、降低MDA含量作用有关。
Objective: To investigate the effect of Naotong compound preparation(NTCP) on erythrocuprein(SOD) activity and malonaldehyde(MDA) content in brains of vascular dementia(VaD) model rats and to explore the preventive and therapeutic effect of Chinese crude drug on spatial learning and memory deficits of VaD rats.Methods: VaD rats were established with modified 4-VO method.These rats were randomly divided into four goups: NTCP treated group(group N),non-treated VaD group(group V),and donepezil hydrochloride treated group(group D).Other 12 SD rats were in sham-operation group(group S).SOD activities and MDA contents in hippocampus of rats in different groups were examined after intervention.Results: After thirty days of intragastric administration,NTCP improved learning and memory abilities of rats in group N.SOD activity increased significantly and MDA level decreased obviously in hippocampus of rats in group N compared with those of rats in group V.Conclusions: NTCP can prevent learning and memory deficit of VaD rats,and the mechanism might relate to increase of SOD activity and decrease of MDA level in brains of VaD rats.
