戴氏绿僵菌提取物对小鼠肿瘤S_(180)抑瘤活性研究

摘要(Abstract):

目的:研究戴氏绿僵菌无性型戴氏绿僵菌培养液上清液中生物活性物质对小鼠肿瘤S180的抗肿瘤活性。方法:体外传代肿瘤细胞株杀伤试验采用MTT法,小鼠体内肿瘤细胞生长检测采用实体瘤称重法,IL-2检测采用酶联免疫吸附测定法(双抗夹心法)。结果:戴氏绿僵菌培养液提取物在高浓度时对体外传代肿瘤细胞株S180有显著的抑制作用,抑瘤率分别为28.06%、23.92%、35.55%、30.92%;对小鼠体内S180实体瘤的生长有显著的抑制作用,抑瘤率分别为22.09%、39.11%、41.67%;能提高荷瘤小鼠血清中IL-2水平,IL-2水平分别为84.627、106.915、136.911ng/L。结论:戴氏绿僵菌提取物在体内和体外均具有抑瘤作用,其抑瘤机理可能与提高机体免疫抑瘤能力有关。
Objective:To investigate the effects of biologically active substances from the culture supernatant of anamorph of Cordyceps taii Liang et liu(SCt)on immunological modulation and anti-tumor activity of the substances to S180 both in vivo and in vitro.Methods:Methyl thiazolyl tetrazolium assay(MTT)was used to detect the ability of killing tumor cell lines.The tumor cell growth in the mice was observed by weighing the weight of tumor.IL-2 in serum was detected with ELISA(sandwich).Results:SCt showed inhibitory effects on the growth of S180 cell in vitro at high concentrations.The inhibitory rates of four concentrations of SCt were 28.06%,23.92%,35.55% and 30.92%.The growth rates of S180 in mice treated with three different concentrations of SCt were smaller than that of mice in negative control group,and the inhibitory rates of the tumor were 22.09%,39.11% and 41.67%.The serum IL-2 levels in tumor-bearing mice treated with the three concentrations of SCt were 84.627,106.915,and 136.911,which were significantly higher than those in mice of both positive and negative control groups.Conclusions:SCt can inhibit the growth of tumor cells both in vivo and in vitro.The mechanism of its anti-tumor effect might be promoting the immunological reaction.
