化疗药物对大肠癌移植瘤裸鼠组织中PCNA蛋白表达的影响Effects of Chemotherapy Drugs on the Expression of PCNA Protein in Transplantation Tumor of Colorectal Carcinoma in Nude Mouse

摘要(Abstract):
目的:探讨化疗药物敏感性与裸鼠皮下大肠癌移植瘤组织中增殖细胞核抗原(PCNA)蛋白表达的关系。方法:用大肠癌患者肿瘤组织建立裸鼠皮下移植瘤模型,分为实验组和对照组;实验组分别予以5-氟尿嘧啶、奥沙利铂、伊立替康、丝裂霉素、阿霉素腹腔注射,对照组大鼠予生理盐水腹腔注射,然后测试各组的肿瘤抑瘤率,采用免疫组化染色方法检测移植瘤组织中PCNA蛋白表达的变化。结果:伊立替康,奥沙利铂,氟尿嘧啶,丝裂霉素,阿霉素组的抑瘤率分别为48.4%,47.8%,40.7%,35.2%和14.5%,平均PCNA阳性细胞计数分别为(38.74±6.16)%、(44.09±4.92)%、(51.56±5.50)%、(56.64±4.57)%、(71.98±4.93)%,对照组为(79.33±3.77)%。结论:化疗药物可降低裸鼠大肠癌移植瘤组织PCNA蛋白的表达强度,化疗药物的敏感性越高,裸鼠皮下移植瘤组织PCNA蛋白的表达越弱。
Objective: To study the relationship between the sensitivity of chemotherapy drugs and the expression of PCNA protein in transplantation tumor of colorectal carcinoma in nude mouse.Methods: The subcutaneous transplantation tumor model of nude mouse was established with colorectal carcinoma,and then rats were randomly divided into 6 groups: 5-fluorouracil group,oxaliplatin group,irinotecan group,mitomycin group,adriamycin group and control group.Rats in control group were given saline instead of chemotherapy drugs.The tumor inhibiting rates were detected and the expression of PCNA protein in tumor tissues was determined with immunohistochemical staining.Results: The tumor inhibiting rates of irinotecan,oxaliplatin,5-fluorouracil,mitomycin and adriamycin were 48.4%,47.8%,40.7%,35.2% and 14.5% respectively,and the average PCNA protein positive expression cell counts were(38.74%±6.16)%,(44.09%±4.92)%,(51.56%±5.50)%,(56.64%±4.57)% and(71.98%±4.93)% respectively.Positive cell count of control group was 79.33±3.77%.Conclusions: The chemotherapy drugs can reduce the expression of PCNA protein in transplantation tumor of colorectal carcinoma,and the higher the susceptibility of the drugs,the weaker the expression of PCNA protein in tumor tissues.
