摘要(Abstract):
目的 :研究肾上腺髓质增生 (AMH)大鼠模型肾上腺组织酪氨酸羟化酶基因表达水平。方法 :Wistar大鼠随机分为 2组 :正常对照组 (NC组 )和肾上腺髓质增生组 (AMH组 )。用利血平皮下注射制备肾上腺髓质增生大鼠模型。 6周后取出肾上腺组织 ,用免疫组织化学和原位杂交法分别测定该模型肾上腺组织酪氨酸羟化酶蛋白质及mRNA表达 ,检测血清中的去甲肾上腺素 (NE)、肾上腺素 (E)和 2 4h尿 3 甲氧 4 羟基苦杏仁酸 (VMA)的含量 ,同时测量肾上腺髓质百分数以反映肾上腺髓质增生的程度。结果 :肾上腺髓质增生大鼠模型肾上腺组织酪氨酸羟化酶蛋白质及mRNA表达均明显增强 ,血清中的NE ,E和 2 4h尿VMA的含量明显升高 ,肾上腺髓质百分数明显升高。结论 :肾上腺髓质增生大鼠模型肾上腺组织酪氨酸羟化酶基因表达增强。
Objective: To investigate the expression of tyrosine hydroxylase (TH) gene in the adrenal of adrenal medulla hyperplasia rat.Methods: Wistar rats were randomly divided into 2 groups: normal control group (NC) and adrenal medulla hyperplasia group (AMH). AMH models were made by injecting of reserpine, and after 6 weeks of the injection, adrenals were taken out from all of the rats to measure the TH protein with immunohistochemical staining and to check the expression of TH gene with in situ hybridization method. At the same time, vein blood was drawn from inferior vena cava to measure the levels of serum NE and E, and 24-hour urine was collected to measure the level of VMA. Adrenal medulla rate was measured to reflect its hyperplasia.Results: The expression of TH gene in group AMH was significantly higher than that in group NC (P<0.01), and the levels of serum NE and E, and 24-hour urine VMA in AMH group were significantly higher than those in group NC (P<0.01).Conclusions: The expression of TH gene increase significantly in adrenals of adrenal medulla hyperplasia rat.
