摘要(Abstract):
目的:评价鼻咽癌患者新辅助化疗前后外周血白细胞中的凋亡相关基因Fas及FasL水平变化及意义,探讨其在鼻咽癌发生、发展及治疗中的作用及临床价值。方法:采用流式细胞仪对30例鼻咽癌患者新辅助化疗前后及10例正常人外周血白细胞中的Fas、FasL的水平进行检测。结果:Fas、FasL在正常人外周血白细胞的表达分别是0和(2.65± 0.15)%,在鼻咽癌患者化疗前外周血白细胞的表达平均为(10.25±2.13%)和(6.97±0.27)%,鼻咽癌组显著高于正常对照组(P<0.01,P<0.05)。鼻咽癌患者经新辅助化疗后,Fas、FasL在外周血白细胞的表达平均为(19.66±3.82)%和(7.35±0.42)%,与化疗前比较Fas水平明显增高(P<0.05),而FasL改变不明显(P>0.05)。结论:外周血白细胞Fas水平对鼻咽癌诊断、评价疗效及预后有一定意义,同时对深入探讨化疗药物对鼻咽癌的治疗有一定基础和临床价值;外周血白细胞的FasL在鼻咽癌发生、发展中起作用,而与鼻咽癌治疗关系不大。
Objective: To evaluate the level change and its significance of relevant apoptosis genes, Fas and FasL, in periphery blood leukocyte of patients with nasopharyngeal carcinoma ( NPC) before and after the neo - adjuvant chemotherapy, and to investigate the function and clinical value of it in occurrence, development and treatment of NPC. Methods: Levels of Fas and FasL in periphery blood leukocyte of 30 patients with NPC before and after the neo - adjuvant chemotherapy and 10 normal people were tested with flow cytometry . Results: The expression of Fas and FasL in periphery blood leukocyte of normal people and patients with NPC were 0, (2. 65± 0. 15) % and (10. 25 ± 2.13)%, (6. 97 ± 0. 27) % respectively. The genes'expression level was much higher in NPC group than that in normal group (P <0. 01, P <0. 05) . For NPC patients received neo - adjuvant chemotherapy, the expression of Fas and FasL in periphery blood leukocyte were (19.66 ±3.82)% and (7.35±0.42)% respectively. The expression level of Fas increased obviously compared with it before the chemotherapy (P<0.05). But, there was no obvious change in expression level of FasL (P > 0. 05). Conclusions : The expression level of Fas in periphery blood leukocyte shows certain significance for the diagnosis, and evaluation of curative effect and prognosis of NPC. And, also, it has certain basic and clinical value for further investigation on the treatment of NPC with chemotherapy. The expression level of FasL in periphery blood leukocyte has some effects in the occurrence and development of NPC, but less relevant with the treatment of NPC. It is showed that flow cytometry test is a convenient and practical way to study the expression of Fas and FasL in periphery blood leukocyte of patients with NPC.
