胰岛素和生长抑素在生后发育期家兔胰腺中的表达
Changes of Insulin and Somatostatin Expression in Rabbit Pancreas during the Postnatal Development Period and Its Significance

摘要(Abstract):

目的:探讨生后发育期家兔胰岛内胰岛素(insulin,Ins)和生长抑素(somatostatin,SST)的表达及分布特点。方法:生后5、25、35、60、90 d家兔共50只,取胰腺组织制成连续切片,用SABC法和图像分析法对Ins和SST免疫反应(immunoreactive,IR)细胞的定位表达进行研究。结果:家兔生后5 d,Ins-IR和SST-IR细胞团数量多染色深;25~90 d,Ins-IR细胞平均灰度值呈升高(P<0.05)又降低的变化,SST-IR细胞生后25 d时平均灰度值最高(P<0.05),此后平均灰度值降低;胰岛内Ins-IR和SST-IR细胞面数密度值(NA)在25 d时最高(P<0.05)。结论:Ins和SST在胰腺中的表达随家兔生长发育而变化,提示Ins和SST参与调节生后发育期家兔胰腺内分泌功能的成熟过程。
Objective: To investigate the expression and distribution features of insulin(Ins) and somatostatin(SST) in rabbit pancreas during the postnatal development period.Methods: Pancreas of rabbits respectively aged 5,25,35,60 and 90 days were taken to prepare serial sections.The localization and expression of Ins and SST in immunoreactive(IR) cells were detected with SABC staining method and image analysis method.Results: Many deeply stained Ins-IR-cell and SST-IR-cell mass were found in 5 days after birth.The mean grey values of Ins-IR cells ascended(P<0.05) and then declined during 25 d to 90 d.The mean grey values of SST-IR cells were highest on the 25th day,and decreased gradually along with the growth of rabbits.NA of Ins-IR cells and SST-IR cells in islets were highest on the 25th day(P<0.05).Conclusions: The expressions of Ins and SST change with the growth of rabbits,which suggests that Ins and SST may participate in the maturation process of pancreas endocrine function during the postnatal developmental period.
