检测抗HSV-1 IgG的斑点金免疫渗滤方法的建立

摘要(Abstract):

目的:建立检测人血清中抗单纯疱疹病毒Ⅰ型IgG(抗HSV-1 IgG)的斑点金免疫渗滤法(D IGFA)。方法:增殖纯化HSV-1抗原并包被于硝酸纤维素膜,自制胶体金并用于标记羊抗人IgG,自制斑点金免疫渗滤检测装置,用于检测血清中抗HSV-1 IgG。结果:当血清样本中抗HSV-1 IgG为阳性时,滴加的金标羊抗人IgG可使检测装置膜上出现红色斑点。以D IGFA与酶联免疫吸附试验(ELISA)用于检测血清样本41份,两法差异无显著性(2χ=0.01,P>0.05)。结论:建立了检测抗HSV-1 IgG的斑点金免疫渗滤方法,该法较其他方法检测时间短,操作简便快捷。
Objective: To establish a simple and rapid method with immunogold filtration assay(DIGFA) for detection of anti-HSV-1 IgG in sera.Method: A nitrocellulose membrane coated with HSV-1 antigen was used to exam anti-HSV-1 IgG in sera.When adding serum sample on the membrane,red dots will appear showing positive ones,since the colloidal gold on the membrane was labeled with goat anti-human IgG.Results: The test results of 41 samples of DIGFA method were not significantly different from those of ELISA(χ2=0.01,P>0.05).Conclusion: DIGFA is a simpler and rapider method for the detection of anti-HSV-1 IgG in sera compared with other methods.
