PI3K/Akt/mTOR信号转导通路在鼻息肉中的表达

摘要(Abstract):

目的:研究磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(Akt)与哺乳动物雷帕霉素靶蛋白(mTOR)在鼻息肉中的表达及相关性,探讨PI3K/Akt/mTOR信号转导通路在鼻息肉发病过程中的作用。方法:鼻息肉组织28例和正常下鼻甲组织12例,用免疫组化Elivison二步法及Western blot检测鼻息肉组与对照组组织中PI3K、Akt与mTOR的表达,比较两组之间的表达差异。结果:鼻息肉组PI3K、Akt与mTOR免疫阳性细胞数及着色强度均明显高于对照组,图像分析显示鼻息肉组PI3K、Akt、mTOR的积分光密度(IOD,103/HP)分别为30.36±3.78、32.70±2.30、35.67±1.26,与对照组3.63±1.55、2.38±1.36、2.79±0.78相比有显著性差异(P值均<0.01);Spearman等级相关分析显示,鼻息肉组PI3K、Akt、mTOR表达密切相关(P<0.01);Western blot检测发现PI3K、Akt与mTOR在鼻息肉组织中特异性表达强度高于对照组(P<0.01)。结论:鼻息肉组织中PI3K、Akt和mTOR表达均上调,三者有协同表达关系,提示可能是通过PI3K/Akt/mTOR信号转导通路使炎性细胞凋亡受到抑制,导致慢性炎症反应,最终形成鼻息肉。
Objective: To study the expression of phosphatidylinositol 3-kinases(PI3K),protein kinase B(Akt) and mammalian rapamycin target protein(mTOR)in nasal polyps and relationship among them,and to explore the role of PI3K/Akt/mTOR signal transduction pathway in the pathogenesis of nasal polyps.Methods: Twenty-eight nasal polyp tissues and 12 normal inferior turbinate mucosa tissues were collected.Immunohistochemistry and western blot were used to detect the expression of PI3K,Akt and mTOR in tissues of nasal polyp group and normal control group,and the expression differences were compared.Results: Immunohistochemistry results shows that the number of positive stained cells and staining intensity of PI3K,Akt and mTOR in nasal polyp tissues were higher than those of controls.Integral optical density(IOD) of PI3K,Akt and mTOR in nasal polyp tissue were higher than those of controls(P<0.01).Spearman correlation test showed that there was a close correlation among PI3K,Akt and mTOR expression in nasal polyp tissues(P<0.01).The specific bands of PI3K,Akt and mTOR in nasal polyp tissues were stronger than those in the control group(P<0.01).Conclusions: The expression of PI3K,Akt and mTOR is up-regulated in nasal polyps,and there is a close correlation among them.
