骨髓间充质干细胞移植治疗大鼠实验性心肌梗死

摘要(Abstract):

目的:研究骨髓间充质干细胞(MMSCs)移植治疗大鼠实验性心肌梗死(MI)远期疗效是否稳定。方法:采用开胸结扎冠状动脉左前降支(LAD)的方法制作MI动物模型,造模成功后12~14 d经舌下静脉移植已转染增强型绿色荧光蛋白(pEGFP-C1)的MMSCs,分4周及8周两时段检测动物的体重变化、血流动力学、组织病理学、微血管密度并用荧光显微镜观察。结果:无论在4周还是8周移植组体重较同期模型组体重显著增加,移植组心率(HR)较同期模型组显著下降。4周时移植组动脉收缩压(SBP)、动脉舒张压(DBP)及平均动脉压(MAP)较模型组无明显改变,8周时则较模型组显著增高。心脏组织病理学改变:4周时移植组较模型组心梗形态学改变减轻,8周时则进一步减轻。荧光显微镜下可见移植组MI边缘区域新生的微血管内皮细胞上有绿色荧光表达;移植组的微血管密度较同期模型组的微血管密度明显增高。结论:MMSCs移植可较为有效地改善大鼠实验性MI后心功能状况,远期疗效也较为稳定。
Objective: To study the long-term therapeutical effect of marrow mesenchymal stem cell(MMSC) transplantation on rats with myocardial infarction(MI) and its stability.Methods: MI model was established by ligation of left anterior descending coronary artery(LAD) in adult SD rats.MMSCs transfected with enhanced green fluorescent protein(pEGFP-C1) were injected into sublingual vein of model rats in 12~14 d afterwards.Body weights,hemodynamic indexes,histopathology of cardia and microvessel density of the rats were detected in 4 weeks and 8 weeks after transplantation.Results: Body weight increased and heart rate(HR) decreased significantly in MMSCs transplantation group(group T) in both 4 weeks and 8 weeks after transplantation compared with those in model group(group M).There was no significant difference of arterial systolic blood pressure(SBP),arterial diastolic blood pressure(DBP) and mean arterial pressure(MAP)between groups T and M in the time point of 4th week.But these indexes in group T increased obviously at the 8th week than those in group M.The morphologic changes in group T at the 4th week were milder than those in group M,and further alleviated at the 8th week.Green fluorescence was detected in microvascular endothelial cells in marginal zone of myocardial infarction of group T under fluorescence microscope.Microvessel density of group T increased obviously than that of group M.Conclusions: MMSCs transplantation can improve heart function of MI rats effectively,and the long-term therapeutical effect is stable to some extent.
