单细胞多重替代扩增法结合比较基因组杂交技术检测植入前发育停滞胚胎染色体非整倍性

摘要(Abstract):
目的:应用单细胞多重替代扩增法(MDA)结合比较基因组杂交(CGH)技术检测植入前发育停滞胚胎染色体非整倍性情况,探讨植入前胚胎发育停滞机制。方法:胚胎植入前对70枚6~8细胞期发育停滞胚胎中取单个卵裂球与正常男性外周血单个淋巴细胞,分别采用多重替代扩增法(MDA)进行全基因组扩增,将发育停滞卵裂球扩增产物标记绿色荧光,淋巴细胞扩增产物标记红色荧光,应用CGH技术进行分析,观察染色体的非整倍性变化情况。结果:对70枚植入前发育停滞的胚胎有7枚出现染色体非整倍性改变,2枚卵裂球染色体为46,XY,dup(Y)(q12);其余5枚卵裂球的染色体分别为45,X;47,XY+21;47,XY+1;46,XY,dup(17)(q23);46,XX,dup(20))(p12);染色体异常的发生率为10%。结论:染色体非整倍性改变可能是植入前胚胎的发育停滞的原因之一。
