TGF-β2和TGFβRⅡ在子痫前期胎盘中的表达Expression and Significance of TGF-β2 and TGFβRⅡ in Placenta of Pre-eclampsia Patients

摘要(Abstract):
目的:检测子痫前期(Pre-eclampsia,PE)胎盘组织中转化生长因子-β2(TGF-β2)和受体TGFβRⅡm RNA和蛋白的表达。方法:正常足月剖宫产孕妇30例为对照组,子痫前期剖宫产孕妇30例(20例足月妊娠、10例未足月妊娠)为子痫前期组,比较两组孕妇的年龄、孕周、胎盘重量及新生儿重量,采用Real-time PCR、Western blot和免疫组织化学方法检测娩出胎盘组织的TGF-β2及TGFβRⅡ的m RNA和蛋白表达。结果:子痫前期孕妇胎盘重量及新生儿体重均低于对照组(P<0.05);子痫前期孕妇胎盘组织中TGF-β2和TGFβRⅡ的m RNA和蛋白表达水平均显著高于对照组孕妇胎盘组织(P<0.05);免疫组化显示子痫前期胎盘中TGF-β2和TGFβRⅡ的阳性率均高于对照组孕妇的胎盘组织(P<0.05)。结论:子痫前期胎盘组织中TGF-β2及TGFβRⅡ升高可能与子痫前期的发生有关。
Objective: To investigate the m RNA and protein expression levels of transforming growth factor beta-2( TGF-β2) and transforming growth factor beta receptor 2( TGFβR Ⅱ) in placentas of pre-eclampsia( PE) patients. Methods: Thirty cases of normal gravidas served as control group and30 cases of PE patients as PE group. Gravidas age,gestational weeks,placenta weight and neonatal weight between the two groups were compared. The m RNA and protein expression levels of TGF-β2and TGFβRⅡ of the specimens were determined by real-time PCR,Western blot and immunohistochemistry. Results: The placenta and neonatal weight of PE group were lower than those of control group( P < 0. 05); TGF-β2 and TGFβR Ⅱ m RNA and protein levels in PE group were significantly higher than those of control group( P < 0. 05); The positive expression rates of TGF-β2 and TGFβRⅡin PE group were significantly higher than those of normal placentas detected by immunohistochemistry method( P < 0. 05). Conclusion: The high expression levels of TGF-β2 and TGFβRⅡ might be related to the aetiology of PE.
